血管紧张素转移酶基因多态性与脑卒中的关系

中文摘要:目的探讨血管紧张素转换酶(ACE)基因插入/缺失(I/D)多态性与北京房山地区汉族人群脑卒中的易感相关性。 方法采用聚合酶链反应(PCR)方法分析63例脑出血患者、713例脑梗死患者和235例正常对照组ACE基因的插入/缺失多态性,观察DD、ID、Ⅱ基因型频率及其等位D、I基因频率,以及其与血糖、血脂水平的联系。 结果房山地区汉族人群脑出血组、脑梗死组和正常对照组相比较,3组的DD、ID、II基因型频率和D、I等位基因频率无显著性差异。ID、II基因型的脑出血患者和脑梗死患者的血糖水平显著高于正常对照组(P<0.01),DD基因型脑梗死患者的血糖水平也高于正常对照组(P<0.05)。DD基因型脑梗死患者的三酰甘油水平、ID型脑出血患者的总胆固醇水平均高于正常对照组,差异有统计学意义(分别为P<001,P<0.05)。结论ACE基因插入/缺失多态性不是脑出血和脑梗死的独立危险因素。但可能影响人血糖、三酰甘油和总胆固醇的水平。

Association Between Angiotension-converting Enzyme and Stroke
Abstract:ObjectiveTo investigate the correlation between ACE gene polymorphisms and stroke of Han nationality people in Fangshan district of Beijing. MethodsThe Insertion/Deletion(ID) polymorphisms of ACE gene were detected in 63 patients with cerebral hemorrhage, and 713 patients with cerebral infarction and 235 health control by polymerase chain reaction(PCR). We observed the frequencies of genotype of deletion homozygote(DD), insertion homozygote(II) and insertion/deletion heterozygote (ID) and the alleles of D and I. Also we analyzed the association among I/D polymorphisms of ACE gene with serum glucose(GLU),triglyceride(TG), cholesterol(TC) levels. ResultsThere was no significant difference in the frequencies of both genotypes of DD,ID,II and alleles of D and I in three groups. The serum GLU levels in patients carrying ID,II genotype were higher than those in healthy control(P<0.01), so were the cerebral infarction patients with DD genotype(P<0.01). As compared to healthy control, the TG levels of cerebral infarction patients with DD genotype and the TC levels of cerebral hemorrhage patients with ID genotype were both higher(P<0.01,P<0.05,respectively). ConclusionACE gene polymorphisms (I/D) is not an independent risk factor for stroke of Han nationality in Fangshan. But it might have association with the serum glucose, triglyceride and cholesterol levels.
