白藜芦醇对缺氧诱导大鼠乳鼠心肌细胞凋亡的作用

中文摘要:目的观察白藜芦醇(Res)对缺氧诱导大鼠乳鼠心肌细胞凋亡的保护作用。方法建立体外培养大鼠乳鼠心肌细胞缺氧模型, MTT法检测心肌细胞活力,Hoechst33258染色检测心肌细胞凋亡,测定心肌细胞内总抗氧化能力(T-AOC)和谷胱甘肽过氧化物酶(GSH-Px)活性。结果经8h、12h、16h、24h缺氧培养后,心肌细胞抑制率分别为(22.13±3.22)%、(29.75±0.34)%、(37.43±6.42)% 和(45.47±7.32)%;心肌细胞缺氧培养24h后,呈现典型的凋亡形态学变化;心肌细胞内GSH-Px活性从(46.96±8.36)U/ml下降到(27.13±4.76) U/ml(P<0.01);心肌细胞内T-AOC为(2.68±0.31)U/ml,缺氧24h后下降到(1.28±0.27)U/ml(P<0.05)。Res(25、50、75μmol/L)可剂量依赖性降低8h、12h、16h、24h缺氧培养所引起的心肌细胞抑制率升高(P<0.05,P<0.01或P<0.001);心肌细胞凋亡形态学改变逐渐改善, 50μmol/L和75μmol/L Res作用更明显;剂量依赖性升高心肌细胞内GSH-Px活性和T-AOC水平(P<0.05 或P<0.01)。 结论Res可减轻缺氧诱导大鼠乳鼠心肌细胞凋亡程度。

Effects of Resveratrol on Neonatal Rat Cardiomyocyte Apoptosis Induced by Hypoxia
Abstract:AbstractObjectiveTo investigate the effect of resveratrol(Res) on neonatal rat cardiomyocyte lesion induced by hypoxia. MethodsThe cardiomyocytes of neonatal rats were cultured in vitro and the model of cardiomyocytes hypoxia was established. The cardiomyocyte vitalities were determined by MTT assay. The cardiomyocyte apoptosis was detected by Hoechst33258 fluorescent staining. The levels of total antioxidant capacity(T-AOC) and glutathione peroxidase(GSH-Px) were measured as well. ResultsAfter the administration of hypoxia for 8, 12, 16 and 24 hours, the cardiomyocyte inhibitory was(22.13±3.22)%, (29.75±0.34)%, (37.43±6.42)% and (45.47±7.32)%, respectively. After the administration of hypoxia for 24 hours, the typical morphologic changes of apoptosis in cardiomyocytes were showed.The intracellular GSH-Px activity decreased from(46.96±8.36)U/ml to(27.13±4.76)U/ml (P<0.01).The level of T-AOC was attenuated from (2.68±0.31)U/ml to(1.28±0.27)U/ml(P<0.05.Res(25,50,75μmol/L)could dose-dependently inhibited the raising of cardiomyocyte inhibitory induced by hypoxia for 8, 12, 16 and 24 hours(P<0.05,P<0.01 or P<0001). The morphologic changes of apoptosis in cardiomyocytes was improved.The effects of 50μmol/L and 75μmol/L Res were more obvious. The levels of GSH-Px and T-AOC of cardiomyocytes increased dose-dependently(P<0.05 or P<0.01).ConclusionRes could improve the apoptosis of neonatal rat cardiomyocyte induced by hypoxia.
