玉米须水提物对糖尿病肾病模型大鼠肾组织转化生长因子β1表达的影响

中文摘要:目的观察玉米须水提物对链脲佐菌素诱导的糖尿病肾病模型大鼠肾组织转化生长因子β1(transforming growth factor-β1,TGF-β1 )表达的影响。方法水溶提取法制备玉米须水提物。 采用高脂高糖饲养加小剂量链脲佐菌素诱导的糖尿病肾病大鼠模型,实验动物分为正常对照组、模型对照组、玉米须水提物干预组,共3组。600mg/(kg·d)剂量的玉米须水提物灌胃给药8周后,测定各组大鼠空腹血糖、体重和肾脏肥大指数,免疫组化法分析肾脏中肾小球内TGF-β1表达水平,并作HE染色观察。结果与正常对照组比较模型组空腹血糖、肾脏肥大指数和肾小球内TGF-β1表达显著升高。 与模型对照组比较,玉米须水提物组可显著降低糖尿病大鼠空腹血糖、肾脏肥大指数、明显使肾小球内TGF-β1表达减少(P<0.05或P<0.01),并对肾脏病理改变有所改善。结论玉米须水提物对糖尿病肾病具有一定肾保护作用及抗肾小球纤维化的作用,其作用机制可能是通过使TGF-β1表达下调而实现的。

Effects of Stigma Maydis Aqueous Extract on the Expression of Transforming Growth Factor-β1 in Renal Tissue of Diabetic Nephropathy Rats
Abstract:ObjectiveTo observe the effects of stigma maydis aqueous extract(SMAE) on the expression of TGF-β1 in the renal tissue of diabetic nephropathy(DN) rats.MethodsDiabetic nephropathy rats were induced by high-fat and high-sugar dietary feeds and low dose streptozotocin(STZ). Rats were randomly divided into normal control group,model control group,and SAME group. After rats were treated for 8 weeks,level of fasting blood glucose(FBG),body weight(BW), and the relative kidney index(ratio between kidney weight and body weight) were respectively assayed. Immunohistocmical study was adopted to analyze the expression of TGF-β1 in acinus renis. HE taining was used to observe morphology of renal tissue. ResultsCompared with Model control group, level of FBG, BW, the relative kidney index and the expression of TGF-β1 was dramatically lower (P<0.05 or P<0.01), meanwhile, renal pathologic changes were also improved in the group. ConclusionSAME may play a role in renal protection and antifibrosis in rats with DN, and its mechanism maybe due to reducing the expression of TGF-β1.
