中文摘要:目的研究血浆中uPA和PAI-1的浓度水平和乳腺癌患者生物学行为相关性,并探讨浓度水平与乳腺癌常见免疫组化指标的相关性。方法利用免疫组化ELISA法检测88例初治乳腺癌患者血浆中uPA和PAI-1的浓度水平,并以39例健康体检者血浆为对照,结合患者的临床病理学资料分析。结果乳腺癌组血浆中uPA和PAI-1水平明显高于健康对照组(P<0.001);乳腺癌血浆中uPA与PAI-1浓度呈正相关(P<0.001);血浆uPA的浓度与乳腺癌的瘤体大小、组织学分级、临床分期、腋窝淋巴结转移分别呈正相关(P<0.05),而与患者年龄、病理类型无关;血浆PAI-1的浓度与乳腺癌的瘤体大小、临床分期、腋窝淋巴结转移分别呈正相关(P<0.05),而与患者年龄、组织学分型、病理类型无关;ER表达阳性乳腺癌患者,血浆uPA及PAI-1浓度值高于ER表达阴性患者(P<0.01);Her-2阳性的乳腺癌患者,血浆中uPA及PAI-1浓度值明显高于Her-2阴性患者(P<0.01)。结论血浆中uPA及PAI-1高表达与乳腺癌的侵袭、转移密切相关;与ER的状态、Her-2表达之间密切相关;对uPA与PAI-1联合检测可能有助于判断乳腺癌的恶性进度及预测其生物学行为。

Relationship between Plasm Urokinase-type Plasminogen Activator (uPA) and Plasminogen Activator Inhibitor (PAI-1) Level and Biological Behaviors in the Breast Cancer Patients
Abstract:ObjectiveThis study is to explore the correlation of the expression levels of uPA and PAI-1 in plasma and the clinicpathological features of breast cancer. MethodsImmunohistochemistry ELISA was used to detected uPA and PAI-1 in plasma of 88 experimental patients and 39 control populations. ResultsPlasma concentration levels of uPA and PAI-1 were significantly higher than those in the normal control group(P<0.001).uPA in Breast cancer patients had positive correlation with PAI-1 in the plasma concentration (P<0.01) .The concentration of uPA in plasma had positive correlation with the tumor size of the breast cancer,histological grade, clinical stage, axillary lymph node metastasis (P<0.05),had negative correlation with the patient′s age, pathological subtype. The concentration of PAI-1 in plasma had positive correlating with breast cancer tumor size, clinical stage, axillary lymph node metastasis (P<0.05),and had negative correlation with age, histological type, pathological subtype.Concentrations of plasma uPA and PAI-1 expression in the breast cancer patients with ER-positive expression were higher than the ER-negative patients (P<0.01).The plasma levels of uPA and PAI-1 were significantly higher in the breast cancer patients with Her-2 positive expressiol than Her-2 negative patients (P<0.01).ConclusionHigh uPA and PAI-1 expression in Breast cancer patients and tumor invasion and metastasis were closely related,and the same to the ER status and Her-2 expression.Combined detection of malignant breast cancer may help to determine the progress and predict their biological behavior.
