中文摘要:目的观察银杏叶提取物(GbE)对肝组织Smad2/3表达的影响,探讨GbE抗肝纤维化的作用机制。方法采用CCl4诱导大鼠肝纤维化,同时分别以200、100、50mg/(kg·d)的GbE进行干预,并设复方鳖甲软肝片为对照组,6周后取肝组织,原位杂交及免疫组化技术分别检测肝组织Smad2/3mRNA及蛋白的表达。结果模型组大鼠肝组织Smad2/3mRNA及蛋白的表达正常组明显增高,应用不同剂量的GbE及复方鳖甲软肝片干预后肝组织Smad2/3mRNA及蛋白较模型组显著下降。结论GbE降低Smad2/3mRNA及蛋白表达可能是其抗肝纤维化作用的重要机制之一。 |

Effects of Ginkgo Biloba Extract on Expression of Smad2/3 in Rats with Liver Fibrosis |

Abstract:ObjectiveTo evaluate the effects of Ginkgo biloba extrat(GbE) on expression of Smad2/3,thus to investigate the possible mechanism of GbE in rats with hepatic fibrosis. MethodsThe liver fibrosis model was induced by CCl4. The rats in administration groups were given with 200mg/kg,100mg/kg,50mg/kg of GbE by daily gavage.At the end of the sixth week, all the rats were sacrificed and livers were obtained. The expression of Smad2/3 was detected by immunohistochemistry. The expression of Smad2/3mRNA in liver was measured by hybridization in situ. ResultsThe expression of Smad2/3mRNA and Smad2/3 protein were significantly reduced after the administration of GbE and Fufang Biejia Ruangan Pian in model group. ConclusionGbE could reduce the transcription and expression of Smad2/3, and inhibit the occurrence and development of CCl4-induced liver fibrosis. |
