小儿睾丸卵黄囊瘤组织原代培养的不同方法评价

中文摘要:目的采用组织培养技术在体外培养取自人体的睾丸卵黄囊瘤组织,比较并建立人睾丸卵黄囊瘤细胞原代培养的理想方法。方法分别采用机械分散法、 酶消化法和植块培养法进行人睾丸卵黄囊瘤细胞的体外原代培养,倒置显微镜及台盼蓝染色观察并比较三者的培养成功率、细胞活力、细胞形态和数量等,免疫组化法鉴定其细胞来源。结果原代培养的成功率、细胞活力与培养方法密切相关,植块培养法成功率高,细胞活力强,优于其他两种培养方法。培养的原代细胞能贴壁生长,AFP染色均呈阳性,表达于胞质,β-HCG染色均阴性,符合睾丸卵黄囊瘤生物学特征。结论植块培养法是一种科学可行的小儿睾丸卵黄囊瘤细胞原代培养的方法,能为进行人睾丸卵黄囊瘤细胞的分离培养纯化及特性研究提供实验基础。

Evaluation of Effect in Primary Culture of Testicular Yolk Sac Tumor by Different Methods in Childhood
Abstract:ObjectiveTo compare and establish an ideal method for the primary culture of testicular yolk sac tumor in childhood in vitro.MethodsThe tissues of testicular yolk sac tumor were separately cultured by three methods:physical disassociation, tissue digestion and tissue explant.The cell culture efficiency,cell vitality and cell morphology were observed and evaluated by phase-contract microscopy and trypan-blue dying methods. The cell source was identified by immunohistochemistry.ResultsMethods applied in primary culture were important factors affecting the subsequent outcome.The successful rate and cell vitality of tissue explant was significantly higher than others. The primary cell of testicular yolk sac tumor was positive for AFP and negative for β-HCG.ConclusionThe tissue explant is an ideal method for the primary culture of testicular yolk sac tumor in childhood in vitro.It may provide a methodological and experimental foundation for isolation, subculture and biological characterization of testicular yolk sac tumor in future.
