石榴皮鞣质对糖尿病大鼠血管保护作用及其机制初探

中文摘要:目的初步探讨石榴皮鞣质(tannin in punica granatum L.)对糖尿病大鼠血管保护作用及其机制。方法采用腹腔注射链脲佐菌素(75mg/kg)的方法建立糖尿病大鼠模型,将成模大鼠随机分为三组:模型对照组、石榴皮鞣质低剂量组、石榴皮鞣质高剂量组,另取正常大鼠作为正常对照组。连续给药21天后,观察大鼠一般生理状况,测定血清葡萄糖(GLU)、三酰甘油(TG)、C反应蛋白(CRP)、一氧化氮(NO)、内皮素-1(ET-1)浓度以及NO/ET-1比值,并分析其相关性。结果与模型对照组比较,低剂量的石榴皮鞣质可显著改善糖尿病大鼠生理状态,显著降低血清GLU、TG、CRP、ET-1浓度,显著提高血清NO浓度、NO/ET-1比值。单因素相关分析显示:血清GLU浓度与ET-1浓度呈显著正相关,与NO/ET-1比值呈显著负相关;血清TG浓度与ET-1浓度呈显著正相关,与NO/ET-1比值呈显著负相关。结论石榴皮鞣质对糖尿病大鼠血管有较好的保护作用,其机制可能与其降低血糖、TG浓度,直接或间接的升高ET-1浓度、降低NO/ET-1比值有关。

Study on Vascular Protection and Its Mechanism of Tannin in Punica Granatum L
Abstract:ObjectiveTo study the vascular protection and its mechanism of tannin in punica granatum L. in diabetes rats. MethodsDiabetic rat models were induced by i.p. injection of streptozotocin (75mg/kg). The diabetic rats were randomly divided into three groups: model control group,tannin in punica granatum L. low-dose group and high-dose group. Normal control group was composed by normal rats. Physiological state of of rats in each group, serum glucose (GLU),triglyceride (TG), Creactive protein (CRP), NO, ET-1 levels, NO/ET-1 ratio and the correlation analysis were studied after 21 days medication administration. ResultsCompared with model control group, low dose of tannin in punica granatum L. can improve the physiological state of diabetic rats significantly, lower serum CRP, GLU, TG and ET-1 levels significantly, and increase serum NO level and NO/ET-1 ratio significantly. Linear regression analysis showed that both the level of serum GLU and serum TG had significant positive correlations with serum ET-1, but negative correlations with NO/ ET-1 ratio. ConclusionTannin in punica granatum L. has the function of vascular protection in diabetic rats. Its mechanism may be related to lowering serum glucose, TG level, directly or indirectly increasing the concentration of ET-1 and reducing NO/ET-1 ratio.
