微囊化卵巢细胞对去卵巢大鼠骨质疏松的影响

中文摘要:目的通过观察去卵巢大鼠微囊化卵巢细胞移植后血清激素及骨代谢相关因子的变化,探讨微囊化卵巢细胞作为防治绝经后妇女骨质疏松方法的可行性。 方法将12周龄SD雌性大鼠30只随机分成3组,每组10只:假手术组(SHAM组)、去卵巢组(OVX组)和微囊化卵巢细胞移植组(MOVT组)。OVX组和MOVT组大鼠切除双侧卵巢,SHAM组大鼠不切除卵巢,仅切除与卵巢大小相当的脂肪组织。卵巢切除术10天后SHAM组、OVX组大鼠腹腔注射0.9% NaCl溶液2ml,MOVT组大鼠则腹腔注射微囊化卵巢细胞2ml。术后4周尾静脉采血,分离血清测定E2、FSH、LH及骨代谢相关因子TGF-β、IL-6、IGF-1。 结果卵巢切除术后,与SHAM组大鼠比较,OVX组大鼠血清E2的水平显著降低(P<0.01),FSH、LH水平则显著升高(P<001);微囊化卵巢细胞移植后,MOVT组大鼠血清E2水平与OVX组比较显著升高(P<0.05);FSH、LH水平均明显降低,LH更加显著(P<0.01)。卵巢切除术后,OVX组大鼠血清TGF-β、IGF-1水平显著下降,IL-6含量则明显升高,与SHAM组比较差异均非常显著(P<0.01);微囊化卵巢细胞移植后,MOVT组大鼠血清TGF-β、IGF-1水平均明显升高,与OVX组大鼠比较分别有非常显著和显著差异(P<0.01、P<0.05),血清IL-6含量降低,与OVX组大鼠比较差异非常显著(P<0.01)。结论微囊化卵巢细胞移植具有预防去卵巢大鼠骨质疏松发生的作用,微囊化卵巢细胞移植作为临床防治绝经后骨质疏松的一种手段具有良好的应用前景。

Effect of Transplanted Microencasulated Ovarian Cells on Osteoparosis in Ovariectomized Rats
Abstract:ObjectiveTo investigate the feasiblity of transplanted microencasulated ovarian cells as a method to prevent osteoparosis after menopause in women. MethodsTotal 30 12-week SD female rats were randomly divided into three groups:sham operation group (SHAM group), model group (OVX group) and microencapsulated ovarian cell transplanted group (MOVT group). Ten days after oophorectomy,in SHAM group, OVX rats were injected 2ml 0.9% NaCl solution, MOVT rats were injected 2ml microencapsulated ovarian cells.Four weeks after operation, serum was collected to determin the E2,FSH,LH level and related factors of bone metabolism such as TGF-β, IL-6, IGF-1 levels. ResultsE2 level and TGF-β,IGF-1 in OVX group significantly decreased 4 weeks after ophorectomy (P<001),whereas FSH,LH and IL-6 levels increased significantly (P<0.01).Compared with OVX group, TGF-β and IGF-1 levels increased in MOVT group 4 weeks after transplantation (P<0.01, P<0.05 respectively),wheras IL-6 decreased significantly (P<0.01). ConclusionTransplantation of microencapsulated ovarian cells in ovariectomized rats can play a preventive role of osteoporosis,and may have a good prospect in clinic.
