中文摘要:目的制备P(St-g-NIPAAm)支架,利用其温敏性在其表面种植3T3细胞,为进一步培养口腔黏膜上皮细胞、治疗角膜缘干细胞缺损奠定基础。方法通过辐照反应将异丙基丙烯酰胺(NIPAAm)接枝在聚苯乙烯(PS)培养皿的膜表面上,制备[ P (St-g-NIPAAm)]支架,使用口腔黏膜上皮细胞在其表面进行体外培养,并对细胞形态进行观察,通过绘制细胞生长曲线和计算克隆形成率来分析细胞的生长特性。结果该支架具有良好的温度敏感性;3T3细胞在其表面成功生长,且具有增殖能力。结论通过辐照反应制备的具有温敏性的P (St-g-NIPAAm)支架,3T3细胞在其表面成功生长,并具有一定的增殖能力。

Abstract:ObjectiveTo prepare the P (St-g-NIPAAm) trestle,and make use of its thermoresponsive to plant 3T3 cells on its surface,in order to lay a foundation for the further study of cultivation of oral epithelial cells,and offer new thoughts to the treatment of limbal stem cell defect.MethodsN-isopropylacrylamide(NIPAAm) was grafted onto the polystyrene cell culture surface by means of irradiation grafting.The oral mucosal epithelial cells were cultured on the surface.The microscope,the cell growth curve and colony formation rate were used to analyze the cell growth characteristics.ResultsCopolymer membrane had a good temperature sensitivity.The 3T3 cells grew well on its surface,and also proliferated a lot during the study.ConclusionThe P (St-g-NIPAAm) trestle has been prepared by the irradiation reaction,on which the 3T3 cells have not only grown well on the surface,but also proliferated a lot.
