实验性非酒精性脂肪肝血清及肝组织瘦素、脂联素的变化及意义

中文摘要:目的探讨瘦素、脂联素在非酒精性脂肪肝病的发生及发展过程中的作用。方法以高脂高胆固醇饲料喂养制作大鼠非酒精性脂肪肝病模型,设立正常对照组和模型6周组、12周组、18周组。测量动物的体重、肝重,常规HE染色观察肝脏病变,酶联免疫法测定血清和肝组织瘦素和脂联素含量。结果病理学观察显示:模型组6周末呈现单纯性脂肪肝,12周末符合脂肪性肝炎表现,18周末呈现肝硬化;与正常对照组相比,模型组血清瘦素含量从6周末开始显著升高,12周末达最高,18周末又回降,但仍高于正常对照组(P<0.05),肝组织炎症活动度(HAI)变化趋势与之相同,且二者呈显著正相关;模型组血清脂联素6周末开始呈显著梯度下降趋势(P<0.05),肝组织羟脯氨酸(Hyp)含量变化趋势与之相反,两者呈显著负相关。结论在非酒精性脂肪肝病发生发展过程中,血清瘦素升高可能加速单纯性脂肪肝向脂脂性肝炎的转化,而血清脂联素稳定可能抑制肝纤维化的进展。

The Leptin and Adiponectin in Blood and Liver Tissue of Nonalcoholic Fatty Liver Disease in Rats
Abstract:ObjectiveTo study change and significance of leptin and adiponectin in blood and liver tissue during nonalcoholic fatty liver disease in rats.MethodsWistar rats were randomly divided into normal control group and mode 6 weeks group,12 weeks group,18 weeks group.Body weight and liver weight of rats were studied.The histological changes were observed by HE.Leptin and adiponectin in blood sample and liver tissue homogenate were detected by ELISA.ResultsThe liver in model group at 6 weeks and 12 weeks and 18 weeks showed typical and notable steatosis,respectively.Leptin content of serum increased significantly from the 8th week,remarkably higher than the model group,and reached pinnacle at the 12th week,but began to decline at the end of the 18th week,The levels of adiponectin was correlated positively with Hyp.The adiponectin declined significantly from the 8th week.The levels of Leptin was correlated negatively with HAI.ConclusionDuring the occurrence and developing of nonalcoholic fatty liver disease,leptin can expedite the developing from NAFL to NASH;and adiponectin can inhibit the developing of hepatic fibrosis.
