经典Wnt信号通路调控对大鼠心肌梗死后心肌愈合的影响

中文摘要:目的探讨经典Wnt信号通路对大鼠心肌梗死后心肌组织愈合的影响。方法建立大鼠急性心肌梗死模型,采用免疫组化和实时荧光定量PCR技术检测经典Wnt信号通路中的两个关键信号分子Wnt-1、β-catenin蛋白及mRNA在对照组(C组)、梗死组(I组)和非梗死组(N组)各时间点心肌组织中的时空表达;同时观察心肌梗死后心肌组织的病理学变化。结果各时间点心肌组织中Wnt-1、β-catenin的表达水平在C组与N组中无显著性差异。I组4天、7天、10天、14天心肌组织中Wnt-1、β-catenin的蛋白表达水平较C组、N组明显增高(P<0.01);I组1天、4天、7天、10天、14天心肌组织中Wnt-1、β-catenin的mRNA表达水平较C组、N组明显增高(P<0.01);且动态变化与心肌梗死后心肌的愈合过程相平行。通过病理形态学与免疫组化分析发现肌纤维母细胞、内皮细胞在发生梗死的大鼠心脏中增殖和迁移的过程中伴随着Wnt-1、β-catenin的高表达。结论经典Wnt信号通路参与心肌梗死后心肌组织的修复和愈合过程;并对肌纤维母细胞和血管内皮细胞的增殖、迁移起一定的调控作用。

Effects of Canonical Wnt Signal Pathway on Rats′ Myocardial Tissue Repair Pocess After Myocardial Infarction
Abstract:ObjectiveTo investigate influences of canonical wnt signal pathway on rats′ myocardial tissue repair pocess after myocardial infarction.MethodsAfter acute myocardial infarction model of rat was set up,immunofluorescence technique and real-time fluorescent quantitative polymerase chain reaction (RQ-PCR) were used to detect the protein and mRNA expressions of both Wnt-1 and β-catenin in rats′ myocardial tissue from group of infarction(group I),group of non-infarction(group N) and control group(group C) at different intervals.At the same time,pathological changes of post-infarction myocardial tissue were also observed.ResultsThere was no obvious difference in expressions of both Wnt-1 and β-catenin in myocardial tissue between group C and N,but both protein expressions in group I were obviously higher than those from either group C or group N at day 4,7,10 and 14(P<0.01),both mRNA expressions in group I were obviously higher than those from either group C or group N at day1,4,7,10 and 14(P<0.01). The dynamic changes of both expressions paralleled with myocardial healing after myocardial infarction.Pathomorphologic and immunohistochemical analysis showed that during proliferation and migration processes of myofibroblast and endothelial cell in myocardial tissue of rats with myocardial infarction,there were incresed expressions of both Wnt-1 and β-catenin.ConclusionCanonical wnt signal pathway participated and regulated the repairing and healing processes of myocardial tissue after myocardial infarction,meantime,regulated the proliferation and migration of myofibroblast and vascular endothelial cell.
