香烟烟雾暴露对大鼠肺组织CCR7和外周血MIP-3β表达的影响

中文摘要:目的研究香烟烟雾暴露对大鼠肺组织CCR7和外周血MIP-3β表达的影响,探讨免疫细胞在吸烟所致气道慢性炎症中的作用。方法30只雄性Wistar大鼠随机分为不吸烟组、吸烟6周组、吸烟12周组;分别用免疫组化半定量法和酶联免疫吸附试验(ELISA)检测各组肺组织CCR7和外周血MIP-3β的含量。结果(1)与不吸烟组(0.173±0.037)比较,吸烟6周组(0.282±0.029) 和吸烟12周组(0.379±0.038)大鼠CCR7的表达明显增加,差异均有统计学意义(P均<0.01);吸烟12周组CCR7表达与吸烟6周组比较有所增加,差异也有统计学意义(P<0.01)。(2)与不吸烟组(77.94±18.59pg/ml)比较,吸烟6周组(162.51±15.66pg/ml)和吸烟12周组(228.34±26.18pg/ml)外周血中MIP-3β含量明显增加,差异均有统计学意义(P均<0.01);吸烟12周组MIP-3β与吸烟6周组比较有所增加,差异也有统计学意义(P<0.01)。(3)肺组织CCR7与外周血MIP-3β的表达呈正相关(r=0.915)。结论吸烟可使大鼠CCR7和MIP-3β的表达水平增高,二者表达呈正相关,提示免疫细胞可能在吸烟所致气道慢性炎症中发挥作用。

The Effect of Cigarette Smoke Exposure on Expression of CCR7 in Lung and MIP-3β in Peripheral Blood of Rats
Abstract:ObjectiveTo explore the effect of cigarette smoke exposure on the expression of CCR7(Chemokine receptor7) in lung and MIP-3β(Macrophage inflammatory protein-3β)in peripheral blood of rats.MethodsThirty male (n=30)Wistar rats were randomly divided into three groups: non-smoking group,smoking 6 weeks group and smoking 12 weeks group.CCR7 was semiquantitatively analyzed in lung by immunohistochemistry.The concentration of MIP-3β in peripheral blood was determined by enzyme-linked immunosorbent assay (ELISA).Results(1)The expression of CCR7 in lung and MIP-3β in peripheral blood of rats in the smoking 6 weeks group (0.282±0.029,162.51±15.66pg/ml)and the smoking 12 weeks(0.379±0.038,228.34±26.18pg/ml) group were higher than those in non-smoking group(0.173±0.037,77.94±18.59pg/ml)(all P<0.01),with the smoking 12 weeks group higher than the smoking 6 weeks group (all P<0.01)(2)The levels of MIP-3β in peripheral blood positively related with CCR7.ConclusionCigarette smoke exposure can upregulate the expression of CCR7 protein in lung and the levels of MIP-3β in peripheral blood in rats,which indicates that immune cells may play an important role in chronic airway inflammation caused by smoking.
