中文摘要:目的本研究旨在探讨红豆杉提取物和纳米乳包裹红豆杉提取物作为裂解灭活H3N2流感疫苗的佐剂对小鼠模型的免疫作用及其毒性程度。方法取ICR小鼠完全随机平均分组后,分别将红豆杉提取物和纳米乳包裹红豆杉提取物作为佐剂与H3N2灭活流感疫苗混合免疫小鼠,同时以相应剂量的单独疫苗组、氢氧化铝佐剂组、生理盐水空白免疫组作为对照组。所有动物于第1周和第3周进行肌内注射免疫,共2次。于初次免疫后每周取血收集血清,测定血凝抑制(HI)抗体效价,并考察每组小鼠的生长状况。同时,进一步观察红豆杉提取物和纳米乳包裹红豆杉提取物的小鼠半数致死量(LD50)。结果红豆杉提取物和纳米乳包裹红豆杉提取物能显著增强血清HI抗体效价,高于单独疫苗组,并且小鼠体重增长正常,结合二者LD50(红豆杉提取物的LD50为295.43mg/kg,纳米乳包裹红豆杉提取物LD50为7.33g/kg)。认为红豆杉提取物和纳米乳包裹红豆杉提取物作为流感疫苗佐剂具有安全有效性,有望成为流感灭活疫苗的新型免疫佐剂。结论本实验首次证实了红豆杉提取物和纳米乳包裹红豆杉提取物可以有效增强对流感抗原的免疫性,并为进一步研究开发提供了毒性依据。鉴于红豆杉提取物和纳米乳包裹红豆杉提取物所表现出的佐剂效应,二者值得做更进一步的研究以便提高疫苗的质量。

Abstract:ObjectiveThis study was designed to evaluate the adjuvants effect and the toxic degree of Taxus madia extracts and Taxus madia extracts-based nanoemulsion on the immune responses elicited by split inactivated H3N2 infiuenza virus antigen in a mouse model. MethodsTwenty-five ICR mice were randomly distributed into five groups with 5 mice in each group. Mice of Taxus madia extracts-adjuvanted inactivated vaccine group and Taxus madia extracts-based nanoemulsion-adjuvanted inactivated vaccine group were given each an intramuscular injection. And mice immunized respectively with inactivated vaccine alone, aluminium-adjuvanted inactivated vaccine and physiological saline which were control groups. All animals were intramuscularly (im.) immunized twice on 0 and 3 weeks. Every week after the boost, blood samples were collected for measurement of HI titers, and the body weight of the five groups were observed. Meanwhile, mouse had been done a further study to observe median lethal dose (LD50) of Taxus madia extracts and Taxus madia extracts-based nanoemulsion. ResultsTaxus madia extracts and Taxus madia extracts-based nanoemulsion significantly enhanced the HI titers, which was higher than those of vaccine alone group, and the body weight grow nomally showed that the both as adjuvents were safe and effective.Consideriring both LD50(the LD50 values of Taxus madia extracts in this experiment were 295.43mg/kg, and the LD50 values of Taxus madia extracts-based nanoemulsion in this experiment was 7.33g/kg), it was reasonable to say Taxus madia extracts and Taxus madia extracts-based nanoemulsion could be used as potential adjuvants of inactivated influenza virus vaccine.ConclusionThis work demonstrates for the first time that Taxus madia extracts and Taxus madia extracts-based nanoemulsion can effectively enhance the immunogenicity of influenza antigen,and provid toxicity data for further research and development.Considering the adjuvants effect demonstrated in this study, both of them deserves further studies for improving the quality of vaccines.
