三七对胰岛素抵抗大鼠血栓形成相关因子的影响

中文摘要:目的研究三七对胰岛素抵抗(IR)大鼠血栓形成相关因子的影响,探讨三七抗胰IR的作用机制。方法32只Wistar大鼠随机分为正常组、模型组、三七高剂量组、三七低剂量组,以高脂饮食4周诱发大鼠IR模型,在造模同时,应用中药三七进行干预,生化法测定空腹血糖(FBG),放免法检测空腹胰岛素(FINS)、TXB2、6-Keto-PGF1α水平,发色底物法测定血浆t-PA、PAI;并计算胰岛素敏感性指标(ISI)。结果IR模型组大鼠的血清FINS较正常组明显增高(P<0.05),而胰岛素敏感性指数ISI明显低于正常组(P<0.05),血浆TXB2、PAI-1水平及TXB2/6-K-PGF1α、PAI-1/tPA比值较正常组明显增高(P<001、P<0.05);应用三七干预后,大鼠FINS水平较模型组明显降低(P<0.05),ISI较模型组明显提高(P<0.05),同时血浆TXB2、PAI-1水平及TXB2/6-K-PGF1α、PAI-1/tPA比值较模型组明显降低(P<0.05)。结论三七明显改善大鼠胰岛素抵抗的作用机制可能与其抑制血栓形成相关因子的表达有关。

Effects of Notoginseng on Thrombosis-associated Factors in Rats with Insulin Resistance
Abstract:ObjectiveTo study the effects of notoginseng on thrombosis-associated factors in rats with insulin resistance to explore the effective mechanism of Notoginseng in the treatment and prevention of IR.MethodsTotally 32 Wistar rats were randomly divided into four groups :the normal group,the control group, the high-dose Notoginseng group and the low-dose Notoginseng group. IR in all rats had been induced by high-fat diet for 4 weeks. Simultaneously, the rats in the treatment groups were intervened with the notoginseng. At the end of fourth week, all rats were put into death,the contents of FBG were measured with biochemical method, the amounts of FINS,TXB2 and 6-Keto-PGF1α with radio immunoassay, the levels of the t-PA,PAI with substrate chromatography and the ISI was calculated. ResultsCompared with rats in the normal group, rats in the control group had more higher FINS (P<0.05),lower ISI (P<0.05) in serum, and had more higher levels of TXB2, PAI-1, TXB2/6-K-PGF1α and PAI-1/tPA (P<0.01,P<0.05), after the treatment with high -dose or low-dose Notoginseng, compared with the model rats, the levels of serum FINS became more lower (P<005),ISI became more higher, and the levels of serum TXB2, PAI-1 and the ratio of TXB2/6-K-PGF1α and PAI-1/tPA became more lower. ConclusionNotoginseng can significantly ameliorate insulin resistance of rats. This mechanism may be associated with the inhibitory effect of Notoginseng from thrombosis-associated factors.
