CACNB2基因多态性与钙离子通道阻滞剂降压疗效的相关性研究

中文摘要:目的探讨钙离子通道辅助β亚基(CACNB2)6个SNP位点多态性对单一钙离子通道阻滞剂(calcium channel blockers,CCB)治疗原发性高血压疗效的影响。方法104例原发性高血压患者同时服用单一钙离子通道阻滞剂(纳欣同)20mg,每日1次,随访6周,采用多重聚合酶链反应(multi-PCR)方法及基质辅助激光解析-飞行时间质谱分析技术(MLD1-TOF MS)对患者CACNB2的6个SNP进行基因分型检测,研究不同SNP位点以及同一SNP不同分型对CCB治疗高血压的疗效差异。结果rs11014166 AA基因型患者服用钙拮抗剂的降压有效率高于AT+TT基因型患者,差异有统计学意义(P<0.05)。其余5个SNP位点不同基因型患者用钙拮抗剂的降压效果差异无统计学意义(P>0.05)。结论CACNB2 rs11014166多态性与钙拮抗剂降压疗效相关,AA基因型人群对钙拮抗剂较为敏感。

Association of Gene Polymorphisms in CACNB2 with the Efficacy of Calcium Channel Blockers in the Treatment of Hypertension
Abstract:ObjectiveTo investigate the association between 6 single nucleotide polymorphisms(SNP) of L-calcium channel auxiliary subunit β2 gene (CACNB2) and the efficacy of calcium channel blockers (CCB) in the treatment of essential hypertension. MethodsA total of 104 EH patients were given 20mg CCB(extended release nifedipine tablets) monotherapy everyday for six weeks.Six SNPs in the CACNB2 determined by multiplex polymerase chain reaction (multi-PCR) and matrix assisted laser desorption ionization time of flight MS (MLD1-TOF MS) to analyze the association of the CCB-efficacy for hypertension with different SNPs and their genotypes. ResultsThe AA genotype of rs11014166 was more efficient than AT and AA genotype(P<0.05).Efficacy of genotypes of other five SNPs was not different each other(P>0.05).ConclusionThe rs11014166 polymorphism of CACNB2 was associated with curative effect of L-calcium channel blockers, which was more effective in patients with AA genotype EH.
