BMP-7在大鼠肝纤维化组织中的动态表达和意义

中文摘要:目的探讨大鼠肝纤维化组织中骨形态发生蛋白7(bone morphogenetic protein-7,BMP-7)的表达趋势及意义。 方法采用腹腔内注射二甲基亚硝胺(DMN)构建大鼠肝纤维化模型,造模后4天、1、2、4、6、8周分别检测血清ALT、AST、ALB的变化,同时取肝组织用半定量RT-PCR方法检测BMP-7 mRNA的表达。采用HE染色及Masson三色染色,光学显微镜下观察肝组织损伤情况。采用单因素方差分析进行多组均数间的比较。 结果肝纤维化模型组血清ALT、AST明显升高,ALB明显下降。BMP-7 mRNA在对照组大鼠和肝纤维化模型组大鼠肝组织中均有表达。与对照组相比,肝纤维化模型组4天时BMP-7 mRNA表达显著减少(P<0.05)。1~2周与对照组差别无显著性(P均>0.05),1周与4天比较差异无显著性(P>0.05),2周与4天比较显著升高(P<0.05)。4周较对照组显著升高,达高峰(P<0.05)。6周较对照组显著升高(P<0.05),较4周下降(P<0.05)。8周较6周有显著下降(P<0.05),与对照组无显著差异(P>0.05)。结论BMP-7 mRNA在正常SD大鼠肝脏有表达,在肝纤维化大鼠肝组织中的表达呈动态改变,在炎症早期表达减少,随着肝纤维化进展,表达增加,在肝纤维化后期表达减少,提示BMP-7参与肝纤维化的发生、发展过程。

Dynamic Expression and Significance of BMP-7 in Liver Tissues of Rats with Hepatic Fibrosis
Abstract:ObjectiveTo explore the expression and significance of bone morphogenetic protein-7(BMP-7) in rats models of hepatic fibrosis. MethodsSD rat models with hepatic fibrosis were established by intraperitoneal injection of dimethyl nitroxide(DMN). Serum alanine aminotransferase(ALT), aspartate aminotransferase(AST) and albumin(ALB) levels were tested from hepatic fibrosis group at the time of 4d, 1week, 2week, 4week, 6week, 8week. And levels of BMP-7 mRNA were detected by semiquantitative RT-PCR in hepatic tissue. Pathological characters of liver tissue were also observed under optical microscope after hematoxylin-eosin and Masson staining. Comparisons between groups were analyzed by one-way ANOVA. ResultsALT and AST in model group were significantly higher than those in control group, but ALB in model group decreased. All rats in normal control group and hepatic fibrosis groups expressed BMP-7. Compared with the rats in normal control group, the expression of BMP-7 mRNA in the 4-day group was significantly lower, but 1/2- week fibrosis groups had no significant difference. Compared with the rats in the 4-day group, the expression of BMP-7 mRNA in the 1-week group had no significant difference, but that of 2-week group was higher obviously. They reached a peak at 4-week, and decreased significantly in the 6-week group in which the expression of BMP-7 mRNA was significantly higher than that of normal control group. The expression of BMP-7 mRNA in the 8-week group was significantly lower than that of 6-week group, but had no significant difference with that of normal control group. ConclusionBMP-7 mRNA expressed in the liver of normal SD rats, and expressed dynamicly in liver fibrogenesis. The expression of BMP-7 mRNA decreased at the early stages of inflammation, and increased along with the progress of hepatic fibrosis, and decreased obviously at the late stage of hepatic fibrosis. These indicated that BMP-7 mRNA might be involed in the pathogenesis and development of hepatic fibrosis.
