中文摘要:目的 研究B淋巴细胞刺激因子受体(BAFF-R)介导的NF-κB信号通路在多发性骨髓瘤(MM)中的作用,并进一步研究NF-кB信号通路对MM细胞存活的影响。 方法 应用Western blot法分析BAFF-R对NF-κB信号通路相关蛋白的激活情况;同时通过RT-PCR、ELISA、WST-1检测NF-κB信号通路抑制剂(BAY11-7082)干预前后BAFF-R mRNA和蛋白表达水平以及对MM细胞增殖的影响。 结果 BAFF-R阻断性抗体(0、1、5和15μg/ml)能够减少p52和p65蛋白的入核,增加IκB-α蛋白表达量,即BAFF-R能够激活NF-κB信号通路。BAY11-7082(1、2和4μmol/L)显著降低BAFF-R mRNA、蛋白表达水平,减少MM细胞增殖和存活。 结论 BAFF-R激活NF-κB信号通路促进多发性骨髓瘤细胞的增殖及存活,进而参与多发性骨髓瘤的发生、发展。

Study on NF-κB Signaling Pathway Mediated by BAFF-R in Multiple Myeloma Cells and Its Effect on Cell Proliferation and Survival
Abstract:Objective To investigate the effect of NF-κB signaling pathway in multiple myeloma cells, and its effect on MM cells survival. Methods Protein levels of NF-κB signaling pathway were detected by Western Blot. BAFF-R mRNA, protein and the proliferation ability of MM cells were examined before and after treated with the inhibitor of NF-κB signaling pathway (BAY11-7082) by real-time PCR, ELISA and WST-1. Results BAFF-R block antibody (0, 1, 5, 15 μg/ml) reduced p52 and p65 protein into the nucleus, increased the expression of IκB-α protein, namely BAFF-R could activate the NF-κB signaling pathway. BAY11-7082(1, 2, 4μmol/L)decreased BAFF-R mRNA, protein and the proliferation ability of MM cells. Conclusion BAFF-R activate NF-κB signaling pathway to promote proliferation and survival of multiple myeloma cells, and then participate in the occurrence of multiple myeloma.
