褪黑素对LPS诱导的肺动脉平滑肌细胞P38MAPK和NF-κB表达的影响

中文摘要:目的 观察褪黑素对LPS诱导的大鼠肺动脉平滑肌细胞(PASMCs)炎症的保护作用及作用机制。 方法 体外培养SD雄性大鼠PASMCs,随机分为5组:空白对照组(N组)、脂多糖(LPS)组、脂多糖+褪黑素(LPS+MLT)组(0.5mmol/L、1.0mmol/L)、LPS+P38MAPK抑制剂(SB203580)(LPS+SB)组(5μmol/L)。CCK-8试验检测褪黑素对PASMCs的药物毒性;半定量RT-PCR分别检测mRNA水平丝裂原活化蛋白激酶(mitogen activated protein kinase, MAPK)通路中P38MAPK和核因子κB(nuclearactor-κB,NF-κB)的表达量;Western blot法检测P38MAPK和NF-κB以及磷酸化P38MAPK和NF-κB的表达量。 结果 在实验设计的浓度内,褪黑素对PASMCs无药物毒性不良反应(P>0.05)。与LPS组相比,LPS+MLT和LPS+SB抑制MAPK信号通路中的磷酸化P38MAPK和磷酸化NF-κB的表达(P<0.01),且随褪黑素的剂量的增加而降低(P<0.01)。 结论 褪黑素抑制MAPK信号通路中P38MAPK和NF-κB的激活,减轻LPS诱导的PASMCs炎性反应。

Effects of Melatonin on the Expression of P38MAPK and NF-κB of LPS in the Pulmonary Artery smooth Muscle Cells Induced by LPS
Abstract:Objective To investigate the effects of melatonin on the expression of P38MAPK and NF-κB in rat pulmonary artery smooth muscle cells (PASMCs) induced by LPS. Methods The cultured male SD rat PASMCs were divided into control group,lipopolysaccharide (LPS) group, melatonin+LPS(MLT+LPS)group (0.5mmol/L,1.0mmol/L) and SB203580+LPS (SB+LPS) group(5μmol/L). The toxicity of melatonin was detected by Cell Counting Kit-8 (CCK-8) assay. The expression of P38MAPK and NF-κB mRNA were detected by RT-PCR. The changes in protein levels of P38MAPK, NF-κB, phosphorylated P38MAPK and phosphorylated NF-κB were detected by Western blot. Results Melatonin showed no toxicity on rat PASMCs in designed concentration (P>0.05). Compared to LPS group, the expression of phosphorylated P38MAPK and phosphorylated NF-κB were down-regulation in MLT+LPS group and SB+LPS group, and in a dose-dependent manner (P<0.01). Conclusion Melatonin can alleviate the LPS-induced inflammation reaction in rat PASMCs by inhibiting the activation of P38MAPK and NF-κB in MAPK signal pathway.
