兔膝骨关节炎模型的建立及其在Dickkopf干预下的病理学改变研究

中文摘要:目的 建立兔膝关节炎模型并探讨Dickkopf-1(Dkk-1)干预下的病理学改变。 方法 构建兔膝关节炎模型,对比观察1个月后对照组、模型组、Dkk-1干预组的病理学表现,并进行Manikin's评分,评估治疗效果。 结果 兔膝关节炎模型构建成功。Dkk-1组虽然未达到对照组正常兔膝关节软骨状态,但使用Dkk-1干预的兔较模型组单纯注射生理盐水的兔膝关节炎病变明显好转。同时,对照组Manikin's评分为0.71±0.40分,模型组为11.93±0.62分,而Dkk-1组为9.02±0.84分。Dkk-1组评分低于模型组,但高于对照组,差异有统计学意义(P=0.005,P=0.007)。 结论 Dkk-1干预可明显改善兔膝关节炎的病理表现和Manikin's评分,但其机制和在人体应用情况尚需进一步研究。

Rabbit Models of Knee Osteoarthritis and the Pathological Characteristics in Dkk-1 Intervention Therapy on Osteoarthritis
Abstract:Objective To build the rabbit models of knee osteoarthritis and study the pathological characteristics in Dkk-1 intervention therapy on osteoarthritis. Methods The rabbit models of knee osteoarthritis were built. After 1 month of treatment of Dkk-1 intervention, the pathological characteristics of control group, model group and Dkk-1 group were contrasted and the Manikin's score was carried out as well. The therapeutic effect was evaluated. Results The rabbit models of knee osteoarthritis were built successfully. The state of the rabbit knee joint cartilage of Dkk-1 group had not got the level of the control group, but the rabbit knee arthritis lesions of Dkk-1 group improved significantly than that of the model group. The Manikin's score of Dkk-1 group(9.02±0.84), lower than the score of model group(11.93±0.62), was significantly higher than that of control group(0.71±0.40). Conclusion Dkk-1 intervention can significantly improve the symptoms of rabbit knee arthritis and Manikin's score, but its mechanism and application in the human body still needs further research.
