离焦诱导豚鼠近视眼的生物学参数及视网膜超微结构特征

中文摘要:目的 观察离焦诱导豚鼠近视眼生物学参数及视网膜超微结构特征。方法 将35只3周龄的三色豚鼠用-10D硬性角膜接触镜(RGP)对右眼进行离焦诱导,左眼不戴镜。3周后比较双眼屈光度,眼轴变化;电子透射显微镜观察视网膜超微结构特征。结果 右眼诱导前后屈光度差值为3.18±0.14D,左眼为1.37±0.13D,二者比较差异有统计学意义(P<0.05);右眼诱导前后眼轴差值为0.62±0.04mm,左眼为0.19±0.02mm,二者比较差异有统计学意义(P<0.05);电镜观察显示3周后诱导眼视网膜可见膜盘的轻度水肿,部分脱落;线粒体肿胀变形;内外核层细胞及色素上皮细胞的少量空泡样改变,细胞膜不规则收缩,核染色质不规则聚集等凋亡特征; 视网膜节细胞未见明显异常改变。结论 -10D的RGP可以成功诱导豚鼠近视眼发生,在近视眼的形成过程中伴随着视网膜细胞的异常凋亡。

Biological Parameters and Retinal Ultrastructural Features of Defocus-induced Myopia in Guinea Pigs
Abstract:Objective To observe the biological parameters and retinal ultrastructural features of defocus induced myopia in guinea pigs. Methods There were 35 guinea pigs (three-week-old) received -10D rigid contact lenses (RGP) for the right eye to be defocus-induced and wihout wearing glasses for the left eye.We compared eye diopter, axial length and observed ultrastructural features of the retina in transmission electron microscopy after 3 weeks. Results Eye diopter difference before and after induction was 3.18±0.14D in right eye and 1.37±0.13D,in left eye.There was significant statistical difference between the two.Axial length difference before and after induction was 0.62±0.04mm in right eye and 0.19±0.02mm in left eye.There was significant statistical difference between the two. Retinal ultrastructural features were showed with transmission electron microscopy(TEM) the retinal disk membranes edema, some shedding, mitochondrial swelling and deformation,small amount of vacuolar changes in the inner and outer nuclear layer cells and pigment epithelial cells.Some characteristics of apoptosis were also be seen such as cell membrane irregular contractions, irregular nuclear chromatin aggregation.But retinal ganglion cells showed no abnormal change after defocus-induction of three weeks. Conclusion The -10D RGP can successfully induce myopia in guinea pigs. Accompanied by abnormal apoptosis of retinal cells during the formation of myopia.
