中文摘要:目的 研究Küppel样转录因子KLF2在急性脑梗死患者外周血单个核细胞中的表达及与血清TNF-α、IL-6浓度的相关性,初步探讨KLF2在急性脑梗死损伤中的可能作用。方法 分别应用半定量反转录-聚合酶链反应(RT-PCR)、双抗体夹心酶联免疫吸附法(ELISA)测定45例急性脑梗死患者及45例健康体检者的外周血单个核细胞KLF2 mRNA表达及血清TNF-α、IL-6浓度,按不同发病时间分为急性期(1~3天)和恢复期组(10~14天),按梗死面积分为腔隙性梗死组(20例)、小面积梗死组(15例)、大面积梗死组(10例)。对各组与对照组之间KLF2 mRNA表达水平及血清TNF-α、IL-6的浓度进行比较。结果 正常对照组KLF2 mRNA呈高表达,急性期及恢复期组低于对照组,差异有统计学意义(P<0.05),急性期KLF2 mRNA表达水平低于恢复期,差异有统计学意义(P<0.05)。急性期TNF-α、IL-6含量均较恢复期增高,恢复期较对照组增高,差异有统计学意义(P<0.05)。脑梗死面积越大,KLF2 mRNA表达水平越低,差异有统计学意义(P<0.05)。结论 脑梗死后KLF2 mRNA表达水平降低,且与血清TNF-α、IL-6的浓度存在相关性,其可能通过介导急性脑梗死的炎性反应在脑梗死发病中有一定的作用。

Expression and Clinical Significance of KLF2 in Peripheral Blood Mononuclear Cells in Patients with Acute Cerebral Infarction
Abstract:Objective To investigate the expression of KLF2 mRNA in peripheral blood mononuclear cells,and its correlation with concentration of TNF-α and IL-6 in patients with acute cerebral infarction,to primarily discuss the role of KLF2 in ischemical injury of acute cerebral infarction. Methods The expression of KLF2 mRNA in peripheral blood mononuclear cells in 45 cases of acute cerebral infarction were examined by real-time quantitative reverse transcription ploymerse chain reaction (RT-PCR). The concentration of TNF-α and IL-6 in serum were examined by enzyme linked immunosorbent assay (ELISA). They were divided into two groups by different periods(1-3 days and 10-14 days)and divided into lacunar CI group (20 cases),small size group(15 cases)and large size group (10 cases).Fourty-five cases of health people were regard as control group. Each group was compared with control group between KLF2 mRNA,TNF-α and IL-6. Results The expression of KLF2 mRNA in control group was higher than acute phase group and recovery phase group.The expression of KLF2 mRNA in recovery phase group was higher than acute phase group,and the difference was significant(P<0.05).The serum levels of TNF-α and IL-6 in acute phase group were higher than recovery phase group,and they in recovery group were also higher than control group,and the difference was significant(P<0.05). The more cerebral infarct size, the lower KLF2 mRNA expression level. Conclusion The expression of KLF2 mRNA decreases. There was negative correlation between with the concentration of TNF-α and IL-6. It possibly plays a role in the pathogenesis of acute cerebral infarction through exerting inflammation injury.
