中文摘要:目的 探讨miR-449b对子宫内膜腺癌细胞HEC-1-B增殖和凋亡的影响及其作用的分子途径。 方法 脂质体lipofectamine 2000包被miR-449b并转染入HEC-1-B细胞,实时荧光定量PCR检测转染前后细胞内miR-449b的表达水平,CCK-8试验检测转染前后细胞活性,流式细胞仪来检测转染前后细胞凋亡情况。细胞克隆形成实验观察转染前后细胞克隆形成能力。Western blot方法检查转染前后细胞内E2F3蛋白和p53的表达情况。 结果 转染后实验组miR-449b表达量较NC组增加,细胞增殖能力和克隆能力减弱,而细胞凋亡增加(P<0.05)。miR-449b使E2F3表达下调,p53表达上调(P<0.05)。 结论 miR-449b通过E2F3-p53途径抑制HEC-1-B细胞增殖并促进凋亡,可能作为治疗子宫内膜腺癌的新分子靶标。

MiR-449b Inhibits Growth of HEC-1-B Cells by Targeting E2F3 and p53 Proteins
Abstract:Objective To investigate the effect of miR-449b on cell proliferation and apoptosis in HEC-1-B cells and the mechanism of such effect. Methods We up-regulated expression of miR-449b in HEC-1-B cells by transfecting them with has-miR-449b.Transfection efficiency was determined by flow cytometry and q-PCR. We then determined the changes in cell proliferation, colony formation and cell apoptosis after transfection. In addition, western blot was applied to determine the target protein of miR-449b. Results Increased-expression of miR-449b significantly inhibits cell proliferation and colony formation and induces cell apoptosis in HEC-1-B cells. The expression of E2F3 protein was down-regulated and the expression of p53 protein was up-regulated. Conclusion MiR-449b inhibits the growth of HEC-1-B cells by targeting E2F3 and p53 proteins.
