中文摘要:目的 研究冠心病患者外周血单个核细胞(peripheral blood mononuclear cells PBMC)中SIRT1 mRNA的表达水平,探讨氨氯地平在冠心病治疗中与SIRT1可能相关的机制。 方法 实验分为3组,阴性对照组、基础用药组、氨氯地平组各21例,分别测药物干预前3组研究对象SIRT1 mRNA表达水平及其与空腹血糖、血脂相关性;服药6个月后,复测两用药组SIRT1 mRNA表达水平及其与空腹血糖、血脂相关性;两用药组药物干预前后对比,研究SIRT1 mRNA表达水平、空腹血糖、血脂变化情况。 结果 ①阴性对照组单个核细胞中SIRT1 mRNA表达水平较基础用药组、氨氯地平组高,差异均具有统计学意义(P<0.05);②药物干预前,基础用药组、氨氯地平组SIRT1 mRNA表达水平差异无统计学意义(P>0.05);药物干预后,氨氯地平组SIRT1 mRNA水平较基础用药组水平高(P<0.05);③药物干预前后对比,基础用药组SIRT1 mRNA表达水平差异无统计学意义(P>0.05);氨氯地平组SIRT1 mRNA表达水平明显升高,但差异无统计学意义(P>0.05);④药物干预前后,研究对象SIRT1 mRNA表达水平均与FBG、TC、TG及LDL-C呈负相关,与HDL-C呈正相关。 结论 SIRT1 mRNA表达在调控血脂、血糖水平及冠心病保护方面或起作用,氨氯地平可能参与SIRT1 mRNA的表达和其生物效应调控过程,延缓动脉粥样硬化进展。

SIRT1 mRNA Expression of the Coronary Heart Disease Patients' Peripheral Blood and Amlodipine's Influence
Abstract:Objective To investigate the expression of SIRT1 mRNA in coronary heart disease patients' peripheral blood mononuclear cells(PBMC),as well as the mechanism of amlodipine in treatment of CHD, which may be related to SIRT1. Methods There were three groups: control group (21 cases),foundation treatment group (21 cases)and Amlodipine group(21 cases).We measured the human PBMC SIRT1 mRNA expression level of three groups respectively before drug intervention, then analysed the relationship between SIRT1 mRNA with fasting blood glucose and lipid. After 6 months' treatment, the SIRT1 mRNA expression level of foundation treatment group and amlodipine group were measured again, as well as the relationship between SIRT1 mRNA with fasting blood glucose and lipid. Moreover, we made a comparison between before and after treatment about the SIRT1 mRNA expression level, fasting blood glucose and lipid. Results 1SIRT1 mRNA expression in control group was higher than foundation treatment group and amlodipine group(P<0.05).2There was no difference in SIRT1 mRNA expression between foundation treatment group and amlodipine group before drug intervention(P>0.05),however, with drug' treatment, amlodipine group expressed SIRT1 higher than foundation treatment group(P<0.05). 3SIRT1 mRNA expression between before and after treatment had no difference in foundation treatment group. Amlodipine group had a increase though P>0.05. 4There was negative correlation between SIRT1 mRNA with FBG, TC, TG and LDL-C and positive correlation between SIRT1 mRNA and HDL-C before drug treatment, as well as after the drug intervention. Conclusion SIRT1 may make effects on the regulation of blood glucose and lipid, also on the protection of coronary disease. Amlodipine could delay the progress of atherosclerosis through SIRT1 mRNA probably.
