应用SELDI-TOF-MS筛选多囊卵巢综合征卵泡液中差异蛋白的研究

中文摘要:目的 探讨多囊卵巢综合征(PCOS)患者与正常排卵妇女卵泡液中差异蛋白质的表达,建立PCOS的卵泡液诊断模型。方法 采用表面增强激光解吸-电离-飞行时间质谱(SELDI-TOF-MS)技术检测24例PCOS患者的卵泡液和20例排卵正常的对照患者卵泡液中的蛋白质,采用支持向量机方法建立PCOS的诊断模型,评估诊断模型的预测效能;采用卡方检验检验PCOS的卵泡液模型间特异性和敏感度的不同。结果 PCOS患者与排卵正常妇女的卵泡液中共检测到89个差异的蛋白质,选取前10个统计学意义的蛋白质峰(P<0.05)合并建立诊断模型,获得5个差异的蛋白质,其中2033和5828质荷比(m/z)两个蛋白质峰在PCOS组卵泡液中的表达低于对照组(P<0.05),其余3个蛋白质峰2873、6452、8137m/z在PCOS组卵泡液中呈现高表达(P<0.05);5个卵泡液差异蛋白质峰组合所构建的诊断模型的特异性为99%,敏感度为97%。结论 PCOS患者的卵泡液与正常排卵妇女卵泡液中存在差异表达的蛋白质,PCOS患者卵泡液中表达的蛋白质的变化可能与PCOS患者的卵泡内卵子发育异常和排卵异常有关。

Detection of Discrepant Proteins in Follicular Fluid of Polycystic Ovary Syndrome and Normal Women Using SELDI-TOF-MS.
Abstract:Objective To detect discrepant specific proteins in follicular fluid from women with and without polycystic ovary syndrome(PCOS) and build diagnostic models. Methods Follicular fluid from women with PCOS(n=24) and women with normal ovulation as control(n=20) were evaluated, with surface-enhanced laser desorption/ionization time-of-flight mass spectrometry(SELDI-TOF-MS)technology. Once biomarkers founded, the diagnostic model was studied and validated by leave-one cross validation. Sensitivity and specificity of different PCOS diagnositic models were evaluated by χ2 test. Results A total of 89 qualified protein peaks were detected in follicular fluid from women with and without PCOS and 10 of them had a significant difference(P<0.05).Combined the first 10 protein peaks to set up the diagnositic models, we found five potential biomarkers with m/z of 2033, 5828, 2873, 6452, 8137 with a specificity of 99% and a sensitivity of 97%. Conclusion There are differentially expressed proteins in follicular fluid from women with or without PCOS, which may be affect the development of ovum and anovulation of PCOS women.
