microRNA-205在人非小细胞肺癌中的表达及临床意义

中文摘要:目的检测人非小细胞肺癌(non small cell lung cancer,NSCLC)组织中microRNA-205表达的变化及分析肺腺癌有淋巴结转移患者组织中microRNA-205与EMT相关标志基因表达的相关性。方法收集2011年12月~2013年10月笔者医院收治的NSCLC患者75例,每例患者另取癌旁组织作为对照,荧光定量PCR法检测肿瘤组织microRNA-205的表达及有淋巴结转移患者肺癌组织中EMT相关基因Slug、Snail、vimentin mRNA的表达情况。结果NSCLC癌组织microRNA-205的表达在<3cm肿瘤患者(19.56±10.81 vs 29.36±10.37)、有淋巴结转移(15.46±7.03 vs 37.98±10.35)或临床分期为Ⅰ~Ⅱ(19.09±9.23 vs 30.21±8.96)患者组织中相对低表达,在肿瘤大小、淋巴结转移或临床分期之间差异具有统计学意义(P<0.05),且29例淋巴结转移肺腺癌患者癌组织microRNA-205与EMT重要转录因子Slug和Snail的表达呈显著负相关(Pearson r=-0.417, P=0.024;Pearson r=-0.388, P=0.038)。结论NSCLC癌组织microRNA-205的表达与淋巴结转移密切相关,可能通过调控Slug和Snail分子参与肺腺癌的侵袭转移过程,将为寻找治疗肺癌转移的策略提供新靶点。

Expression and Clinical Significance of MicroRNA-205 in Human Non-small Cell Lung Cancer.
Abstract:Objective To understand the microRNA-205 expression in patients with non-small cell lung cancer (NSCLC), and analysis the correlation of microRNA-205 with EMTassociated gene expression in lung adenocarcinoma patients with lymph node metastasis. Methods Seventy-five NSCLC patients in the hospital from Dec. 2011 to Otc. 2013 were involved in this study. The adjacent noncancer tissues were served as control. Quantitative polymerase chain reaction was used to detect the expression of microRNA-205 in primary NSCLC and expression of three other EMTassociated markers Slug, Snail and vimentin in lung adenocarcinoma with lymph node metastasis. Results microRNA-205 had a relative low expression levels in patients with tumor size less than 3 cm (19.56±10.81 vs 29.36±10.37), lymph node metastasis (15.46±7.03 vs 37.98±10.35) or clinical stages in Ⅰ-Ⅱ (19.09±9.23 vs 30.21±8.96). In NSCLC patients, the expression levels of microRNA-205 exhibited distinct difference among patients at different tumor size, lymph node metastasis and clinical stages (P<0.05), and had a significant negative correlation with the expression of Slug and Snail in patients with lung adenocarcinoma, important transcription factors of EMT (Pearson r=-0.417, P=0.024;Pearson r =-0.388, P=0.038). Conclusion The expression of microRNA-205 was closely associated with lymph node metastasis in NSCLC, and may through regulation the molecular of Slug and Snail to be involved in the process of invasion and metastasis in lung adenocarcinoma. microRNA-205 may provide a new target in seeking treatment for NSCLC metastasis strategy.
