姜黄素对实验性大鼠非酒精性脂肪肝病氧化应激、炎性因子及细胞凋亡水平的影响

中文摘要:目的 探讨姜黄素对实验性大鼠非酒精性脂肪肝病肝脏氧化应激、炎性因子及细胞凋亡等水平的影响并初步探讨其机制.方法 将大鼠随机平均分为正常对照组(NC组)、高脂饮食组(HF组)和高脂饮食+姜黄素治疗组(HF+curcumin组),各组给予相应处理造模后,观察肝脏病理变化并分析相关血清学指标.结果 NC组、HF组和HF+curcumin组大鼠血GSH-Px、T-AOC、SOD分别为28.5±2.5、21.7±3.1、24.1±1.2;4.3±0.9、1.8±1.1、2.9±0.7;163.7±13.4、123.5±21.1、143.1±16.7,MAD分别为1.3±0.5、3.2±0.8、2.4±0.6,血IL-6、TNF-α分别为67.5±12.4、114.5±15.2、91.3±16.1;32.8±5.5、40.5±4.2、34.5±6.1,组间比较差异均有统计学意义(P<0.05或P<0.01);各组间NF-κB/GAPDH及Bcl-2/Bax比值差异均有统计学意义(P<0.05或P<0.01).结论 姜黄素能够减轻NAFLD大鼠肝脏脂肪变性程度,改善胰岛素抵抗、肝功能及病理损伤,明显减轻其发病关键环节氧化应激,炎性反应及细胞凋亡的程度.

Curcumin's Effect on the Level of Oxidative Stress,Inflammation Factors and Apoptosis in Experimental Non-alcoholic Fatty Liver Disease Rats
Abstract:Objective To study the curcumin's effect on the level of oxidative stress, inflammatory factors and apoptosis in experimental nonalcoholic fatty liver disease rats and explore the mechanism preliminarily. Methods All the rats were randomly divided into normal control group (NC group), high fat diet group (HF) and high-fat diet+curcumin treatment group (HF+curcumin group), and each group was given the corresponding processing, to observe liver pathological changes and analyze related serological indexes. Results Rats'blood GSH-Px,T-AOC,SOD in NC group, HF group and HF + curcumin group respectively where 28.5±2.5,1.7±3.1,24.1±1.2;4.3±0.9,1.8±1.1,2.9±0.7;163.7±13.4,123.5±21.1,143.1±16.7 and MAD respectively was 1.3±0.5,3.2±0.8,2.4±0.6,and IL-6,TNF-α respectively was 67.5±12.4,114.5±15.2,91.3±16.1;32.8±5.5,40.5±4.2,34.5±6.1. The differences between groups were statistically significant (P<0.05 or P<0.01). The NF-κB/GAPDH及Bcl-2/Bax ratio in each group is different with statistical significance (P<0.05 or P<0.01). Conclusion Curcumin can improve the hepatic steatosis, insulin resistant(IR), liver function, pathological damage and especially can significantly reduce the key courses as oxidative stress, inflammation and apoptosis in NAFLD rats.
