| 中文摘要:目的 探究lncRNA PCA3高表达在前列腺癌细胞中的功能。方法 将构建的含有PCA3全长的pcDNA3.1质粒及对照瞬时转染PC3细胞，提取细胞总RNA进行反转录，并采用实时定量PCR检测转染效率；采用MTS的方法检测PCA3高表达对细胞增殖能力的影响；利用Transwell实验检测PCA3高表达对细胞迁移能力的影响；采用流式细胞仪检测PCA3高表达对细胞凋亡的影响。结果 高表达后lncRNA PCA3、PC3细胞的增殖、迁移能力增强，UV诱导的凋亡降低。结论 lncRNA PCA3高表达可以促进PC3细胞的恶性表型。 |

| Overexpression of LncRNA PCA3 Promotes Cell Proliferation and Migration Abilities and Inhibits Apoptosis in Prostate Cancer |

| Abstract:Objective To exmamin the functions of overexpression lncRNA PCA3 in prostate cancer cells. Methods After transfection with pcDNA3.1-PCA3 and the control plasmids to PC3 cells, the transfection rates were examined by real time-PCR. MTS assay was applied to evaluate the influence of overexpression on cell proliferation. Transwell assay was used to test the effect on cell migration. Flow cytomet was applied to detect the influence on cell apoptosis. Results The experiments proved that elvated expression of lncRNA PCA3 might accelerate the proliferation ability, promote the migration and decline the apoptosis rate in PC3 cells. Conclusion Overexpression of lncRNA PCA3 promotes the malignancy of PC3 cells. |
