全脑照射对大鼠海马区Notch 1信号通路的影响

中文摘要:目的 观察大鼠全脑照射后海马区Notch 1信号通路中Notch 1与下游因子Hes 1的表达变化。方法 单次10Gy全脑照射构建大鼠放射性脑损伤模型,于照射后1、3天、1、2周、1、2个月处死大鼠获取海马组织,采用real-time PCR和Western blot法检测海马组织Notch 1、Hes 1的表达变化,免疫组织化学染色法检测海马区新生神经细胞数。结果 相较对照组,大鼠全脑照射后Notch 1表达进行性下调;其下游因子Hes 1在照射后2周内表达无明显变化,而在照射1、2个月后表达显著下调(P<0.05)。在照射后1、2个月组中,海马区新生神经细胞数较对照组显著减少。结论 大鼠全脑照射后,海马区Notch 1信号通路活性显著下调,伴随着海马区神经再生显著抑制,提示Notch 1通路可能参与放射性脑损伤后神经再生的调控过程。

Influence of Whole Brain Irradiation on Notch 1 Signaling Pathway in Rat Hippocampus
Abstract:Objective To investigate whether whole brain irradiation could influence Notch 1 signaling pathway in rat hippocampus. Methods Female SD adult rats were suffered by a single dose of 10Gy X-ray whole brain radiation. Then, rats were sacrificed at 1 day, 3 days, 1 week, 2 weeks, 1 month and 2 months post radiation and hippocampuses were isolated. Real-time PCR and Western blot were performed to detect the expression change of Notch 1 signaling pathway. Immunohistochemistry staining was administrated to detect Newborn neurons in hippocampus by using doublecortin(DCX) as the marker. Results Compared to normal control group, the expression of Notch1 showed a tendency of progressive decrease at 1 day, 3 days, 1 week, 2 weeks, 1 month and 2 months after whole brain irradiation at hippocampus. Hes 1, a downstream target of Notch 1 signaling, stayed stable in 2 weeks post irradiation, and resulted in a significant reduction in expression at 1 month and 2 months post whole brain irradiation(P<0.05). Compared to normal control group, the number of newborn neurons was significantly reduced at 1 and 2 months after whole brain irradiation. Conclusion The expression of Notch 1 signaling was significantly downregulated post whole brain irradiation, accompanied by reduction of neurogenesis, which should be involved in the neurogenesis after whole brain irradiation in adult hippocampus.
