高糖活化小鼠小胶质细胞BV-2模型的建立

中文摘要:目的 观察高糖(Glu)对小鼠神经小胶质细胞BV-2活化的影响。方法 将BV-2细胞分成:正常对照组(25mmol/L Glu),脂多糖组(1μg/ml LPS),高糖组(75mmol/L Glu),培养24h,倒置相差显微镜观察细胞形态,MTT检测细胞活力,激光扫描共聚焦显微技术、Western blot法及实时荧光定量PCR技术检测BV-2活化的标志物CD11b的表达。结果 高糖组、LPS组细胞活力与正常对照组相比差异均无统计学意义(P >0.05);高糖组BV-2 CD11b蛋白含量较正常对照组显著升高(P<0.01),与LPS组类似(P >0.05);高糖组BV-2 CD11b-mRNA值较正常对照组显著升高(P<0.01),且显著高于LPS组(P<0.01)。结论 75mmol/L高糖培养BV-2 24h,在不影响细胞活力的条件下,可显著上调BV-2活化标志物CD11b蛋白及mRNA表达,作用与LPS相当。

Establishment of a High Glucose-induced Microglia Activation Model
Abstract:Objective To observe the effect of high glucose on the activation of microglia BV-2 cells. Methods The BV-2 cells incubated in vitro were divided into three groups:control group(25mmol/L Glu), LPS group(1μg/ml),high glucose group(75mmol/L Glu), After being cultured for 24h under different conditions, the morphological changes of microglia BV-2 cells was observed by inverted phase contrast microscope, MTT colorimetric assay was adopted to measure BV-2 proliferation, confocal laser scanning microscopy, western blot and real-time fluorescence quantitative PCR were explored to determine the expression of surface markers CD11b of activated microglia. Results MTT colorimetric assay:After 24h, compared with control group, the OD values of high glucose group, LPS group and different NFC groups showed no significant differences(P >0.05).The expression of CD11b in the Glu group increased significantly compared with those of control group (P<0.01),which showed no statistical difference comparing with LPS positive group(P >0.05). The level of CD11b mRNA transcription in the Glu group were much higher than those of the Con group and LPS positive group. Conclusion After stimulation with high glucose (75mmol/L) for 24h, the transcription and expression of surface expression of activation marker CD1lb in microglia BV-2 cells were noticeably up-regulated, while the vitality of microglia BV-2 cells was not influenced.
