中文摘要:目的 通过实验研究下调miR-205的表达对Ⅱ型子宫内膜癌细胞增殖、侵袭力的影响及对裸鼠皮下移植瘤生长的抑制作用,以探讨将miR-205作为Ⅱ型子宫内膜癌的靶向治疗的潜在靶点的可能性。方法 利用脂质体Lipofectamine 2000将miR-205抑制物转染入Ⅱ型子宫内膜癌细胞HEC-1-B中;利用Q-RT-PCR检测转染后各组细胞中miR-205的表达情况;CCK-8检测细胞增殖能力变化情况;Transwell实验检测穿膜细胞数变化情况;利用BALB/c免疫缺陷裸鼠建立Ⅱ型子宫内膜癌皮下移植瘤模型,并测量各组移植瘤的瘤体重量。结果 Q-RT-PCR结果显示,转染miR-205抑制物组细胞中的miR-205表达水平显著低于无关序列组和空白对照组(P<0.01);下调miR-205表达量对HEC-1-B细胞的增殖和侵袭力均有抑制作用,并能显著降低裸鼠移植瘤的瘤体体积及重量(P<0.01)。结论 在Ⅱ型子宫内膜癌中,miR-205表现出癌基因的生物学行为,下调miR-205的表达可以显著降低Ⅱ型子宫内膜癌HEC-1-B细胞的增殖侵袭能力,miR-205的过表达可能参与了Ⅱ型子宫内膜癌的发生、发展及侵袭转移过程,miR-205将有望成为Ⅱ型子宫内膜癌的生物靶向治疗的新靶点。

Regulation of miR-205 in Type Ⅱ Endometrial Carcinoma Biological Behavior
Abstract:Objective To explore the role played by miR-205 in the regulation of type Ⅱ endometrial carcinoma cell proliferation,invasion and tumor growth in nude mice,which may could apply in the biological targeted therapy of type Ⅱ endometrial carcinoma. Methods We downregulated miR-205 in type Ⅱ endometrial carcinoma cell line HEC-1-B by using lipofectamine 2000. The expression of miR-205 after transfection was detected using quantitative real-time PCR. The ability of cell proliferation was studied by CCK-8 and invasion by Transwell.Form xenograft model of type Ⅱ endometrial carcinoma in BALB/c immunodeficient nude mice, and measure tumor xenografts weight of each group. Results MiR-205 was greatly downregulated in transfected group. (P<0.01).The inhibitory action of miR-205 downexpression was confirmed by CCK-8(P<0.01).And there was significant difference in the invasion ability too(P<0.01). Tumor xenografts weight of miR-205 inhibit group was lower than the other two groups. Conclusion Our study suggested that miR-205 may act as a oncogene in type Ⅱ endometrial carcinoma. Downexpression of miR-205 inhibits proliferation and invasion in HEC-1-B cell line,which may be the new target of gene therapy of type Ⅱ endometrial carcinoma.
