中文摘要:目的 检测自噬相关蛋白Beclin1及LC3在良性前列腺增生、前列腺癌标本及盆腔淋巴结中的表达并探讨其意义。方法 应用免疫组织化学方法,检测自噬相关蛋白Beclin1及LC3蛋白在41例前列腺癌组织、60例良性前列腺增生组织,17例转移淋巴结组织及24例正常盆腔淋巴结组织的表达。结果 免疫组织化学方法检测Beclin 1、LC3在正常前列腺增生组织中阳性表达率明显高于前列腺癌组织(P<0.05),不同Gleason分级间前列腺癌的Beclin l、LC3表达差异无统计学意义(P >0.05)。晚期前列腺癌(C+D期) Beclin1、LC3表达水平高于早期(A+B期)。Beclin 1、LC3在转移淋巴结组织中阳性表达率高于正常盆腔淋巴结组织(P<0.05)。结论 自噬相关蛋白Beclin1、LC3参与前列腺癌的发生及发展,在不同临床分期发挥不同作用,细胞自噬抑制剂与化疗药物结合有望成为一个新的治疗靶点。

Expresseion of Beclin1 and LC3 in Prostate Cancer and Relationship with Lymph Node Metastasis
Abstract:Objective To investigate the expression of Beclin 1,LC3 in benign prostatic hyperplasia tissue,prostate cancer tissue and Pelvic lymph node and to explore the significance between it. Methods Immunohistochemistry was used to confirm the expressions of autophagy-related protein Beclin1,LC3 in 60 cases of BPH, 41 cases of prostate cancer,17 cases of metastatic lymph nodes and 24 cases of normal pelvic lymph nodes. Results We found that Beclin l,LC3 expression was higher in BPH tissues than that in Prostate cancer tissues(P<0.05),and havd no statistics difference in different Gleason stage of prostate cancer(P >0.05).The expression of Beclin1,LC3 was higher in the tissues from advancd stages (C+D) than from the early stages (A+B).The expression level of Beclin1,LC3 in metastatic lymph nodes was significantly higher than that in normal pelvic lymph nodes(P<0.05). Conclusion Autophagy-related protein Beclin1,LC3 might be involved in the genesis and development of Pca and play different roles in different clinical stages.Cell autophagy inhibitors and chemotherapy drugs are expected to become a new therapeutic target.
