山楂叶总黄酮对2型糖尿病大鼠睾丸组织的保护作用及其机制研究

中文摘要:目的 研究山楂叶总黄酮(hawthorn leaves favonoids,HLF)对2型糖尿病大鼠睾丸组织的保护作用,并探讨其可能的作用机制。方法 将80只大鼠采用数字表法随机分为正常组、模型组、HLF低(50mg/kg)、中(100mg/kg)、高(200mg/kg)剂量组,每组16只;采用高脂高糖饮食8周后腹腔注射链脲佐菌素(STZ)的方法制备2型糖尿病大鼠模型;灌胃给药,每天1次,疗程6周。治疗完成后,测定各组大鼠空腹血糖水平,称量体重并计算睾丸指数,测定血清睾酮含量;HE染色观察睾丸组织形态结构改变,透射电镜观察睾丸组织超微结构改变;TUNEL染色法观察细胞凋亡状况并计算凋亡指数(apoptosis index,AI)。结果 与模型组比较,HLF中、高剂量组大鼠空腹血糖水平显著降低,体重及睾丸指数显著升高,血清睾酮含量显著升高,差异均有统计学意义(P<0.05,P<0.01)。HE染色后光镜下可见模型组大鼠睾丸曲细精管萎缩、生精细胞脱落且数量显著减少等病理性改变;电镜下可见模型组睾丸组织细胞呈现线粒体减少、内质网扩张、细胞核固缩、染色质呈块状浓集等超微结构改变;模型组睾丸组织凋亡细胞数量较正常组明显增多;经HLF治疗后上述病理性变化和细胞凋亡状况均明显改善,其中以HLF高剂量组效果最为显著且HLF中、高剂量组AI显著降低(P<0.05,P<0.01)。结论 HLF能够有效降低2型糖尿病大鼠空腹血糖,对睾丸的相关保护可能是源自降低血糖带来的收益。

Protective Effects and Mechanism of Hawthorn Leaves Favonoids on Testis Tissue in Type 2 Diabetic Rats
Abstract:Objective To investigate the protective effects and mechanism of hawthorn leaves favonoids(HLF) on testis tissue in type 2 diabetic rats. Methods Eighty rats were randomly devided into five groups: normal group, model group, HLF low(50mg/kg), medium(100mg/kg), high(200mg/kg) dose group,n=16. The diabetic rat models were made by dietting with high-fat diet for 8 weeks and injecting STZ; the drugs were given by intragastric administration for 6 weeks, once a day. The level of plasma glucose was determined. The body weight was detected and the testis tissue index was calculated. The content of testosterone in serum was determined. The histopathological changes of testis tissue was observed by HE staining. The ultrastructure of testis tissue was observed by electron microscope. The apoptosis of testis tissue apoptosis was observed by TUNEL and the AI was calculted. Results Compared with model group, the level of plasma glucose in HLF medium and high dose groups were significantly decreased; while the body weight, the testis tissue index and the content of testosterone were significantly increased. All of the difference above were significant (P<0.05, P<0.01). The seminiferous tubules in the rats of model groups were atrophied, germ cells were significant reduced. And the mitochondrial reduced, endoplasmic reticulums were expanded, lipid droplets were increased, cell nuclear pyknosis, chromatin gathered. The symptoms were significantly improved in HLF high dose group. The testis cells apoptosis were significantly improved and the AI of medium, high dose group were significantly decreased (P<0.01). Conclusion HLF could effectively reduce fasting blood glucose in type 2 diabetic rats, which perhaps was the reason of protective effects on testis tissue injury.
