中文摘要:目的 探讨A型钾通道5种亚型即Kv1.4、Kv3.4、Kv4.1、Kv4.2和Kv4.3在正常大鼠臂旁外侧核(lateral parabrachial nucleus,LPB)的表达。方法 分别采用RT-PCR和实时荧光定量PCR技术检测分布于成年雄性SD大鼠LPBA型钾通道亚型。结果 用RT-PCR技术在所有6只大鼠LPB都检测到了Kv3.4、Kv4.2和Kv4.3 mRNA,但在1只大鼠LPB未检测到Kv1.4 mRNA,2只大鼠LPB未检测到Kv4.1 mRNA;实时荧光定量PCR实验显示,在LPB,Kv1.4、Kv3.4、Kv4.2和Kv4.3 mRNA表达显著高于Kv4.1 mRNA,而Kv3.4、Kv4.2、Kv4.3和Kv1.4 mRNA的表达丰度差异无统计学意义(P>0.05),但在1只大鼠LPB未检测到Kv1.4 mRNA。结论 在正常大鼠LPB中有A型钾通道mRNA表达,其主要亚型为Kv3.4、Kv4.2和Kv4.3。

mRNA Expressions of A-type Potassium Channel in Rat Lateral Parabrachial Nucleus
Abstract:Objective To investigate the mRNA expressions of A-type potassium channel subtypes in rat lateral parabrachial nucleus (LPB). Methods The mRNA levels of Kv1.4, Kv3.4, Kv4.1, Kv4.2 and Kv4.3 were respectively detected with RT-PCR and fluorescent quantitative real time PCR. Results There were mRNA expressions of Kv3.4, Kv4.2 and Kv4.3 in the LPB of all the tested rats by RT-PCR, while Kv1.4 and Kv4.1 mRNA was not detected in the LPB of 1/6 and 2/6 rats, respectively. With fluorescent quantitative real time PCR, the mRNAs of Kv1.4, Kv3.4, Kv4.2 and Kv4.3 were significantly abundant than that of Kv4.1, while the mRNAs of Kv1.4, Kv3.4, Kv4.2 and Kv4.3 did not show difference, although Kv1.4 mRNA was not detected in the LPB in one of the 6 rats. Conclusion The mRNA expressions of A-type potassium channel are detected in rat LPB, where the major subtypes of A-type potassium channels are Kv3.4、Kv4.2 and Kv4.3.
