中文摘要:目的 比较不同产地神曲对小鼠RAW264.7单核-吞噬细胞及大鼠小肠上皮细胞株IEC-6细胞活性的影响。方法 不同产地神曲样品甲醇提取,氮吹仪下挥干溶剂,加入空白培养基配成1g/ml母液,无菌条件下加DMEM空白培养基使母液稀释成浓度分别为800、400、200、100、0μg/ml,对大鼠小肠上皮细胞株IEC-6细胞及小鼠RAW264.7单核-吞噬细胞进行给药培养,检测二者增殖率、巨噬细胞吞噬能力及其所分泌TNF-α的水平。结果 不同神曲样品在低给药浓度(100、200μg/ml)时均有促进IEC-6细胞及RAW264.7细胞增殖的作用,尤其以中国药材公司采集的神曲炒品(Z炒神曲)效果较显著(P<0.05)。对于巨噬细胞吞噬能力及其所分泌的TNF-α水平,各组间虽差异无统计学意义,但仍以Z炒神曲在较低浓度时优于其他组别。结论 不同产地神曲对小肠上皮IEC-6细胞及巨噬细胞RAW264.7活性有促进作用,以从中国中药公司采集的Z炒品效果较优。 |

Effects of Methanol Extracts of Different Habitat Massa Medicata Fermentata(MMF) on the Activity of Macrophages and Small Intestinal Epithelial Cells |

Abstract:Objective To compare the effect of different habitat Massa Medicata Fermentata on the activity of the mouse mononuclear macrophages RAW264.7 and rat small intestinal epithelial IEC-6 cells. Methods Different MMF samples wera extracted with methanol drying solvent with nitrogen blowing instrument,1g/ml initial liquid was made with blank culture medium in a aseptic condition,the initial liquid was diluted into 800,400,200,100,0μg/ml with DMEM blank culture medium to culture mouse mononuclear macrophages RAW264.7 and rat small intestinal epithelial IEC-6 cells or determine the both appreciation rate,macrophage phagocytic ability and the levels of TNF-α. Results Different MMF samples had the effect of promoting the proliferation of IEC-6 cells and RAW264.7 cells at low concentration(100,200μg/ml,which especially remarkable was fried product collected from Chinese traditional Chinese medicine company(Z fried MMF)(P<0.05).Although there was no significant difference between the groups for the phagocytic ability of macrophages and its levels of TNF-α, Z fried product was better than other groups at low concentrations. Conclusion Different MMF samples have a promoting effect on the activity of intestinal epithelial IEC-6 cells and macrophage RAW264.7 cells.Z fried product collected from Chinese traditional Chinese medicine company is better. |
