KDM3A加重高胰岛素诱导的血管平滑肌细胞损伤

中文摘要:目的 探究组蛋白去甲基化酶KDM3A对高胰岛素条件下平滑肌细胞增殖、迁移功能以及炎性状态的影响。方法 从SD大鼠胸主动脉分离原代血管平滑肌细胞(vascular smooth muscle cells,VSMCs),之后将VSMCs随机分为3组∶①正常培养基+对照组干扰siRNA组;②高胰岛素刺激+KDM3A特异siRNA组;③高胰岛素刺激+对照组干扰siRNA组。分别用CCK-8、Transwell检测细胞的增殖和迁移。RT-PCR检测白介素6(IL-6)、单核细胞趋化蛋白1(MCP-1)的mRNA的表达水平。蛋白免疫印记法检测KDM3A的表达。结果 高胰岛素刺激可以显著促进VSMCs的增殖和迁移,同时上调KDM3A的表达,并提高IL-6及MCP-1的mRNA水平。然而,使用siRNA下调KDM3A的表达可以显著减少高胰岛素诱导的VSMCs的增殖、迁移及炎性因子的表达。结论 KDM3A加重高胰岛素诱导的血管平滑肌细胞的损伤。

Histone Demethylase KDM3A Aggravates Proliferation, Migration and the Expression of Inflammatory Cytokine of Vascular Smooth Muscle Cells Induced by High Concentrate Insulin
Abstract:Objective To explore the effect of KDM3A on the proliferation, migration and inflammation state of VSMCs under the circumstance of high concentrate insulin. Methods Primary VSMCs were isolated from thoracic aorta of Sprague-Dawley rats. VSMCs were randomly divided into 3 groups:(1)VSMCs cultured with normal medium+ control siRNA group. (2)High concentrate insulin stimulate+ control siRNA group.(3)High concentrate insulin stimulate+ KDM3A siRNA group. Cellular proliferation and migration were measured by CCK-8, transwell chamber, respectively. The mRNA levels of interleukin-6 (IL-6) and monocyte chemotactic protein-1 (MCP-1) were measured by RT-PCR. Protein expression levels of KDM3A was evaluated by western blotting analysis. Results The proliferation and migration of VSMCs were promoted by the stimulation of high concentrate insulin. At the same time, the protein level of KDM3A and mRNA expression of IL-6 and MCP-1 were increased. However, down-regulate the expression of KDM3A with special siRNA could significantly ameliorate the proliferation, migration and inflammatory cytokine expression of VSMCs induced by high concentrate insulin. Conclusion KDM3A aggravates the injury of VSMC induced by high concentrate insulin.
