中文摘要:目的 研究上皮-间质转化(epithelial-to-mesenchymal transitions,EMT)中主要相关因素上皮细胞钙黏蛋白(E-cadherin,E-cad)和Slug在口腔黏膜下纤维性变(oral submucous fibrous,OSF)组织中的作用研究。方法 采用荧光定量PCR法(Q-PCR)及免疫组织化学法检测41例OSF患者和19例正常口腔黏膜组织切片中E-cad和Slug的表达,分析E-cad与Slug的表达在OSF和正常口腔黏膜组织中的异常表达及E-cad与Slug的异常表达的相互作用关系。结果 免疫组化结果显示在OSF组织中E-cad和Slug的阳性表达率分别为70.73%(29/41)和41.46%(17/41),同时发现OSF组织中E-cad的表达与Slug的表达为负相关(r=-0.656,P<0.05)。荧光定量PCR结果显示OSF组织中Slug的表达高于正常口腔黏膜组织(P<0.05),而OSF组织中E-cad的表达低于正常口腔黏膜组织(P<0.05)。结论 EMT在OSF形成及恶变中可能起重要的作用,其主要相关基因E-cad与Slug的检测对预防OSF恶变成口腔癌提供重要参考指标。

Epithelial-Memsenchymal Transitions Effects on Oral Submucoous Fibrous
Abstract:Objective To study the effects of slug and E-cadherin (E-cad), the mainly relative element in the Epithelial-Memsenchymal Transitions, on the oral submucous fibrous(OSF). Methods We detected the expressions of E-cad and Slug of 41 OSF patients and 19 normal oral mucosal biopsies by adopting the fluorescene quantitative polymerase chain reaction assay (Q-PCR) and immunohistochemical method; analyzed the expressions of E-cad and slug in OSF patients and the abnormal expressions of E-cad and slug in normal oral mucosa, and analyzed the relationship between the abnormal expressions of E-cad and slug and interreaction. Results Immunohistochemical analysis showed that the positive expression rates of E-cad and slug in OSF musoca were respectively 70.73%(29/41) and 41.46%(17/41), and it also showed a negative relation between the expression of E-cad and the expression of slug in OSF musoca. The Q-PCR analysis showed that the expression of slug in OSF musoca was higher than in normal oral musoca (P<0.05), but the expression of E-cad in OSF musoca is lower than in normal oral musoca (P<0.05). Conclusion EMT probably plays an important role in the formation and canceration of OSF, and the detection of mainly relative genes, E-cad and slug can provide important reference index for preventing OSF from cancerating to oral cancinoma.
