淫羊藿女贞子合布地奈德对哮喘大鼠肺组织JAKs/STAT4-5的影响

中文摘要:目的 研究中药淫羊藿女贞子煎剂及提取物配伍对布地奈德气道雾化的哮喘大鼠肺组织JAKs/STAT4-5蛋白磷酸化表达的影响。方法 SPF级雄性SD大鼠,采用数字表法随机分为7组,分别是正常组、模型组、布地奈德组、煎剂组、布地奈德合煎剂组、提取物组、布地奈德合提取物组。卵蛋白致敏、激发建立大鼠哮喘模型,雾化吸入布地奈德和(或)灌胃淫羊藿女贞子煎剂或提取物,免疫荧光法检测肺组织JAKs (包括JAK1、JAK2、JAK3、TYK2)及STAT4、STAT5蛋白的磷酸化表达。结果 与正常组比较,模型组大鼠肺组织pJAK1、pJAK3、pTYK2和pSTAT5蛋白表达均显著上调(P均<0.01),pJAK2和pSTAT4显著下调(P均<0.01)。与模型组比较,5个给药组均能显著影响pJAKs各蛋白的表达(P<0.01或P<0.05),上调pSTAT4蛋白表达的阳性面积,下调pSTAT5蛋白的阳性面积及积分光密度(IOD)(P<0.01或P<0.05)。与单用布地奈德比较,布地奈德合煎剂组或合提取物组对pJAKs蛋白表达和pSTAT4、pSTAT5蛋白表达的Area作用更优(P均<0.01)。结论 在布地奈德治疗哮喘的同时合用中药淫羊藿女贞子,可通过调节JAK/STAT4-5蛋白的磷酸化表达产生协同增效作用。

Effects of Epimeium and Fructus Ligustrilucidi on JAKs/STAT4-5 in Lung Tissue of Asthmatic Rats with Intervention by Budesonide
Abstract:Objective To study the effects of the decoction and extracts of Epimeium and Fructus Ligustrilucidi (EF) on JAKs/STAT4-5 protein phosphorylation in lung tissues of asthmatic rats atomized by budesonide. Methods The healthy male SD rats were randomly divided into seven groups, including normal group, model group, budesonide group, EF decoction group, EF decoction plus budesonide group, EF extracts group, and EF extracts plus budesonide group. Asthmatic Rats model was duplicated by the injection and atomization inhalation of OVA. Atomization inhalation of budesonide and/or gavage EF decoction or EF extracts and the phosphorylation protein expressions of JAKs(including JAK1,JAK2,JAK3,TYK2) and STAT4,STAT5 of lung tissues were detected in rats of all groups by the method of immunofluorescence. Results Compared with the normal group, the protein expression of pJAK1, pJAK3, pTYK2 and pSTAT5 in lung tissues significantly went up (P<0.01), and the protein expression of pJAK2 and pSTAT4 significantly went down (P<0.01). Compared with the model group, the protein expression of pJAKs in five treatment groups significantly changed (P<0.01 or 0.05). The positive area of pSTAT4 significantly went up, and the positive area and SumDensity of STAT5 in lung tissues significantly went down (P<0.01 or 0.05).Compared with the budesonide group, the EF decoction plus budesonide group or EF extracts plus budesonide group had a better effect on the protein expression of positive area of pJAKs, pSTAT4 and pSTAT5. Conclusion Combination of budesonide with EF may produce a synergistic interaction effect on asthma by adjusting JAKs/STAT4-5 protein phosphorylation expression.
