中文摘要:目的 探讨褪黑素在柯萨奇病毒B3(Coxsackie virus B3,CVB3)感染的病毒性心肌炎小鼠中的保护作用,为病毒性心肌炎的治疗提供新的手段。方法 将64只5周龄的雄性BALB/c小鼠随机分为3 组:正常对照组、实验对照组、褪黑素组。以腹腔注射CVB3建立急性病毒性心肌炎模型,接种当天记为第0 天,24h后正常对照组、实验对照组每日分别经腹腔注射等量生理盐水,褪黑素组每日经腹腔注射溶于乙醇及生理盐水的褪黑素。分别于接种后第7天和第14天从各组随机抽取8只处死,留取心肌组织标本。用苏木素-伊红染色法观察心肌炎症病理改变,蛋白印迹法定量检测Bax、Bcl-2、Cleaved-caspase 9的蛋白水平表达,用免疫组织化学法检测心肌组织中Cleaved-caspase 3的表达量。结果 与正常对照组比较,实验对照组心肌炎症病理评分较高,Bax、Cleaved-caspase 9凋亡蛋白表达水平和Cleaved-caspase 3表达量上调,Bcl-2蛋白水平表达下调,给予褪黑素干预后心肌炎症病理评分降低,并可明显下调Bax、Cleaved-caspase 9凋亡蛋白表达水平和Cleaved-caspase 3表达量,上调Bcl-2蛋白表达水平。结论 褪黑素能够减轻心肌损伤,下调病毒性心肌炎小鼠心肌组织中Bax、Cleaved-caspase 3、Cleaved-caspase 9的表达和上调Bcl-2的表达,从而对病毒性心肌炎小鼠起到保护作用。

Protective Role of Melatonin in a Murine Model of Viral Myocarditis Induced by Coxsackievirus B3
Abstract:Objective To investigate the protective role of melatonin in a mouse modle of viral myocarditis induced by Coxsackievirus B3(CVB3). Methods A total of 64 five-week-old BALB/c male mice were divided into three groups:the normal control group, the experimental control group, the melatonin group[14.4g/(kg·d)]. The model of acute viral myocarditis was induced by intraperitoneal injection of CVB3. Inoculation on the same day was labled for 0 day. 24h after the normal control group, the normal control group and experimental control group daily were established by intraperitoneal injection the same amount of normal saline. The melatonin group daily was established by intraperitoneal injection melatonin which were dissolved in alcohol and normal saline, respectively. On day 7 and day 14, heart specimens which were used for evaluation of myocardial edema and inflammation were collected from 8 mice of each group. The myocardial inflammatory pathological changes were observed by using hematoxylin-eosin staining. The protein expression levels of Bax, Bcl-2 and Cleaved-caspase 9 were detected by Western blot. The expression levels of Cleaved-caspase 3 were measured by immunohistochemistry method. Results Compared with the normal control group, inflammation pathological score of the experimental control group was higher, the apoptosis protein expression levels of Bax, Cleaved-caspase 9 were up-regulated, as well as the level of Cleaved-caspase 3 in immunohistochemistry method, the Bcl-2 protein expression levels were down-regulated. With the treatment of melatonin, myocardial inflammatory infiltration was alleviated, the protein expression levels of Bax, Cleaved-caspase 9 were down-regulated, accompanied by the lower level of Cleaved-caspase 3, the Bcl-2 protein expression levels were up-regulated. Conclusion Melatonin can reduce myocardial injury and the expression of Bax, Cleaved caspase 3 and Cleaved caspase 9 in myocardial tissue. At the same time, it can increase the expression of Bcl-2, namely, Melatonin has a protective effect on viral myocarditis.
