中文摘要:目的 观察黄芪甲苷(astragaloside IV,ASIV)对苯并芘(benzopyrene,Bap)诱导RAW264.7巨噬细胞MMPs表达的保护作用,探讨腹主动脉瘤防治的可能新策略。方法 应用Western blot法观察Bap对RAW264.7巨噬细胞基质金属蛋白酶-9/12(MMP-9、MMP-12)、NF-κB、TNF-α等表达的变化及黄芪甲苷的作用,利用流式细胞仪观察黄芪甲苷对巨噬细胞ROS表达的影响。结果 Bap能够诱导RAW264.7巨噬细胞MMP-9、MMP-12等金属蛋白酶的表达升高和NF-κB、TNF-α等炎性指标的活化。黄芪甲苷具有降低RAW264.7巨噬细胞因Bap诱导而表达升高的MMP-9、MMP-12、NF-κB、TNF-α,同时具有清除ROS的作用。结论 黄芪甲苷能够降低Bap刺激RAW264.7巨噬细胞引起的MMP-9、MMP-12、NF-κB、TNF-α的表达升高,并具有清除ROS的作用。

Benzopyrene Induced the Expression of MMPs and the Effects of Astragaloside in RAW264.7 Cells
Abstract:Objective To explore the effect of astragaloside IV (ASIV) in Bap induced the expression of MMPs on RAW264.7 cells, to provide a new control strategy of abdominal aortic aneurysm caused by smoking. Methods We observed the influence of ASIV on matrix metalloproteinase-9/12 (MMP-9,MMP-12) and NF-κB,TNF-α of RAW264.7 cells in vitro by western blotting. We used flow cytometry instrument to analysis whether ASIV has the role of reducing ROS. Results Bap caused expression of MMP-9, MMP-12, NF-κB, TNF-α increased in cultured RAW264.7 cells. ASIV has the effect of decreasing the expression of MMP-9, MMP-12, NF-κB, TNF-α, otherwise it can eliminate the ROS which was caused by Bap incultured RAW264.7 cells. Conclusion ASIV can decrease the expression of MMP-9,MMP-12, NF-κB,TNF-α and eliminate the ROS in RAW264.7 cells cultured with Bap.
