中文摘要:目的 研究岩大戟内酯4种活性单体对人脑胶质瘤U251细胞增殖抑制及诱导凋亡的作用。方法 用不同浓度的岩大戟内酯A (JA)、17-羟-岩大戟内酯A (HJA)、岩大戟内酯B (JB)、17-羟-岩大戟内酯B (HJB)处理U251细胞,分别在12、24、48h时间点,用MTT法检测细胞增殖抑制率;在24h时间点,通过LDH释放检测细胞毒性,流式细胞术检测细胞早期凋亡率,Western blot法检测凋亡相关信号蛋白Bax、Bcl-2、cleaved-caspase-3的表达。结果 JA、HJA对U251细胞无作用。与空白对照组比较,JB、HJB使细胞增殖均受到抑制,并呈浓度和时间依赖关系(P<0.05);使LDH释放率、早期凋亡率、Bax和cleaved-caspase-3表达均随着给药浓度的升高而增大,而Bcl-2的表达降低(P<0.05)。结论 JB和HJB可能通过诱导细胞凋亡而对人脑胶质瘤U251细胞产生作用。 |

Effect of Active Monomers of Jolkinolide on Proliferation and Apoptosis of U251 Cells |

Abstract:Objective To study the effect of active monomers of Jolkinolide on activity of U251 cells.Methods The U251 cells were dealt with different concentrations of Jolkinolide A(JA). 17-Hydroxy-jolkinolide A(HJA), Jolkinolide B(JB), 17-Hydroxy-jolkinolide B(HJB) at 12h, 24h and 48h, cell proliferation inhibition rate was measured by MTT. Cytotoxicity was measured by LDH release. Annexin V-FITC was used to test the apoptosis rate. Expression of apoptosis-related protein bax, bcl-2, Cleaved-caspase-3 was measured by Western blot at 24h.Results JA and HJA had no effect on U251 cells. Compared with normal group, the U251 cells were dealt with JB and HJB. Cells proliferation was inhibited in a time and dose-dependent manner(P<0.05). The release rate of LDH, early apoptosis rate, expression of bax and Cleaved-caspase-3 was advanced with dose-dependent manner, but the expression of bcl-2 was decreased(P<0.05).Conclusion JB and HJB may affect human glioma u251 cells by inducing apoptosis. |
