松果菊苷对2型糖尿病小鼠血脂代谢的影响

中文摘要:目的 探讨松果菊苷对2型糖尿病小鼠血脂代谢影响的特点,并初步探讨其对db/db糖尿病小鼠肝脏组织中成纤维细胞因子21(fibroblast growth factor 21,FGF21)含量的影响,寻找松果菊苷调节2型糖尿病血脂代谢可能的途径。方法 取6周db/db小鼠20只,随机选取10只作为模型组,剩余10只作为实验组,另取6周db/m小鼠8只(SPF级)作为空白对照组,适应2周后检测空腹血糖均>16.7mmol/L进入实验。各组予以灭菌蒸馏水、松果菊苷、灭菌蒸馏水灌喂8周后,取血清测总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、高密度脂蛋白(high density lipoprotein,HDL-C)、低密度脂蛋白(low density lipoprotein,LDL-C)、丙氨酸氨基转移酶(alanine aminotransferase,ALT)、天门冬氨酸氨基转移酶(aspartate aminotransferase,AST)的水平,并取肝脏组织测FGF21的水平。结果 模型组血清TG、TC及LDL含量明显高于对照组(P<0.01),血清HDL的含量明显低于对照组(P<0.01);实验组血清TG、TC及LDL的水平明显低于模型组(P<0.01),血清HDL的含量明显高于模型组(P<0.01);模型组小鼠肝脏中FGF21表达的水平显著低于对照组(P<0.01),实验组小鼠肝脏中FGF21表达的水平显著高于模型组(P<0.01);模型组小鼠血清ALT、AST的含量显著高于对照组(P<0.01),实验组小鼠血清ALT、AST的含量显著低于实验组(P<0.01)。结论 松果菊苷给药8周后能够显著改善db/db糖尿病小鼠血脂代谢紊乱,且不加重肝功能损害,其作用机制可能与其提高肝脏组织中FGF21表达水平有关。

Effect of Echinacoside on Lipid Metabolism in Type 2 Diabetic Mice
Abstract:Objective To explore the characteristics of the effect of echinacoside on lipid metabolism in type 2 diabetic mice, and to investigate the content of growth factor 21 in liver tissue of diabetic mice, providing further insight into the possible way of regulating blood lipid metabolism in type 2 diabetic mice with Echinacea. Methods Twenty db/db mice (6-week-old) were randomly divided into two groups:experimental group and model group; while six db/m mice (SPF) (6-week-old) were control group. We detected the fasting blood glucose after 2 weeks that was higher than 16.7mmol/L in experiment. Each group was given sterilized distilled water, echinacoside, sterilized distilled water. The serum total, cholesterol, triglyceride, high density lipoprotein, low density lipoprotein, alanine aminotransferase level, aspartate, aminotransferase level, and the liver tissue measurement of fibroblast growth factor 21 levels were detected after eight weeks. Results The model group serum TG, TC and LDL were significantly higher than the control group (P<0.01). The content of serum HDL was significantly lower than the control group (P<0.01). Experimental group of serum TG, TC and LDL levels were significantly lower than the model group (P<0.01).The content of serum HDL was significantly higher than that in the model group (P<0.01). The expression of FGF21 in model group in liver were significantly lower than that of control group (P<0.01). The experimental group the expression of FGF21 in mouse liver was significantly higher than that in the model group (P<0.01). The content of ALT and AST in serum of model group was significantly higher than the control group (P<0.01). Experimental group serum ALT, AST were significantly lower than the experimental group (P<0.01). Conclusion After 8 weeks of administration, the serum lipid metabolism disorder of db/db diabetic mice was significantly improved, and the liver function damage was not increased. The mechanism of FGF21 may be related to the improvement of the expression level of liver tissue.
