Caveoline-1蛋白在慢性神经痛大鼠背根神经节和脊髓背角表达的变化

中文摘要:目的 观察Caveolin-1蛋白在神经痛大鼠中不同作用时间脊髓背角和背根神经节表达量的变化。方法 实验动物分组和动物模型制作:对照组28只,神经痛模型组(L5脊神经结扎)28只,按处理时间不同又各分为1、3、5、7、10、14、21天共7组,每组各4只,制备动物模型。痛行为学检测:术后1、3、5、7、10、14、21天,测定各组机械撤足阈值和热撤足潜伏期,神经痛模型组和对照组对比,确定疼痛模型制作成功。神经痛模型组和对照组大鼠均在第22天处死,按同侧和对侧分别提取L5脊髓和背根神经节组织总蛋白,利用蛋白印迹法检测Caveolin-1在各实验分组脊髓背角和背根神经节同侧和对侧的表达量。结果 同侧背根神经节Caveolin-1的表达量在L5脊神经结扎后的第5、7、10、14、21天与对照组比较,分别升高了40.4%±3.67%、126.9%±5.46%、159.7%±4.89%、119.1%±5.77%和91.4%±5.31%,其中术后第10天差异有统计学意义(P<0.01),同侧脊髓背角Caveolin-1的表达量在L5脊神经结扎后的第5、7、10、14、21天与对照组相比分别升高了33.3%±4.89%、152.8%±3.56%、142.1%±5.43%、103.2%±6.21%、175.6%±4.81%,其中第10、14、21天差异有统计学意义(P<0.05)。对照组之间Caveolin-1的表达无明显变化。结论 慢性神经痛可诱导Caveolin-1在同侧脊髓背角和背根神经节表达量的增加。

Expression of Caveolin-1 in Dorsal Root Ganglia and Spinal Dorsal Horn in the Development of Chronic Pain
Abstract:Objective To observe variation of Caveolin-1 expression in dorsal root ganglia and spinal dorsal horn in chronic neuropathic pain of rats. Methods Experimental animals of 56 SD rats were used for this study,and 28 rats for control.They were divided into 7 groups and their L5 nerves were ligated to produce the neuropathic pain model. After 1,3,5,7,10,14,21d of surgery, the mechanical shrinkage foot threshold and hot shrinkage foot incubation period were detected. Neuropathic pain models were confirmed to make successfully compared with the control group. Rats in the surgery group and the control group rats were all put to death on the 22d. The total protein of L5 spinal cord and dorsal root ganglia was extracted from the same side or the opposite side. The Caveolin-1 expression of different groups and sides was detected by the Western blot method in dorsal root ganglia and spinal dorsal horn in chronic neuropathic pain of rats. Results After 5,7,10,14,21d of L5 ligation, the expressions of Caveolin-1 were increased by 40.4%±3.67%,126.9%±5.46%,159.7%±4.89%,119.1%±5.77% and 91.4%±5.31% in the same sides. The difference was significant on 10d(P<0.01) after the surgery. After 5d,7d,10d,14d,21d of L5 ligation, the expressions of Caveolin-1 were increased by 33.3%±4.89%,152.8%±3.56%, 142.1%±5.43%,103.2%±6.21%,175.6%±4.81% in the opposite sides. It had statistic difference on 10d, 14d, 21d(P<0.05). There were no statistic differences among control groups. Conclusion The Neuropathic chronic pain can increase the Caveolin-1 expressions on the same side of dorsal root ganglia and spinal dorsal horn.
