牙周炎大鼠MMP-2、BGP及炎性细胞因子的表达特点及低氧对于相关因子的影响

中文摘要:目的 探讨牙周炎大鼠牙周组织中基质金属蛋白酶(MMP-2)、骨钙素(BGP)、炎性细胞因子的表达及低氧对牙周炎及相关因子表达的影响。方法 选取健康雄性SD大鼠48只,采用数字表法随机分为正常组(正常大鼠,常规氧压下喂养)、牙周炎组(建立牙周炎动物模型)、低氧组(低氧压条件下喂养)、联合组(建立牙周炎动物模型+低氧压)各12只,各组大鼠均在相应条件下喂养8周,对比两组的牙龈指数(GI)、牙周附着丧失(AL)、采用Western blot法检测牙周组织中MMP-2、BGP蛋白表达,采用免疫组化染色法检测白细胞介素-6(IL-6)、白细胞介素-18(IL-18)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)蛋白表达。结果 牙周炎组、低氧组及联合组的牙周组织GI、AL、IL-6、IL-18、TNF-α、IL-1β水平显著的高于正常组,差异有统计学意义(P<0.05);联合组的GI、AL、IL-6、IL-18、TNF-α、IL-1β水平显著的高于牙周炎组、低氧组,差异有统计学意义(P<0.05);牙周炎组、低氧组及联合组的牙周组织MMP-2蛋白水平显著的高于正常组(P<0.05),牙周炎组、低氧组及联合组的牙周组织BGP蛋白表达水平低于正常组(P<0.05);联合组的MMP-2蛋白显著的高于牙周炎组、低氧组(P<0.05),联合组的牙周组织中BGP蛋白水平显著的低于牙周炎组、低氧组(P<0.05)。结论 牙周炎大鼠牙周组织中MMP-2、炎性因子表达增加,BGP表达显著的降低,低氧条件将促进MMP-2、炎性因子表达增加,BGP表达显著的降低从而加重牙周炎病情。

Effect of Hypoxia on Periodontitis and the Effect of MMP-2, BGP and Inflammatory Cytokines
Abstract:Objective To investigate the effects of hypoxia and periodontitis on matrix metalloproteinases (MMP-2), osteocalcin (BGP) and inflammatory cytokines in periodontal tissues of rats. Methods We selected 48 healthy male SD rats.They were randomly divided into normal group (normal rats, normal oxygen periodontitis group (FED), to establish the animal model of periodontitis), hypoxia group (low pressure conditions, the combined group (FED) to establish the animal model of periodontitis + oxygen pressure) with 12 rats in each group. The rats were fed for 8 weeks in the corresponding conditions gingival index (GI), periodontal attachment loss (AL), the expression of MMP-2 and BGP protein in periodontal tissue in the two groups was detected by Western blot assay. Immunohistochemical staining was used to detect interleukin -6 (IL-6), interleukin -18 (IL-18), tumor necrosis factor alpha (TNF-alpha), interleukin -1 beta (IL-1 beta) protein expression. Results The levels of GI, AL, IL-6, IL-18, TNF-α and IL-1β in periodontal tissues were significantly higher than those in normal group (P<0.05). The levels of GI, AL, IL-6, IL-18, TNF-α and IL-1β in the combined group were significantly higher than those in the periodontitis group and hypoxia group (P<0.05). The levels of MMP-2 protein in periodontal tissues of periodontitis group, hypoxia group and combined group were significantly higher than those in normal group (P<0.05). The expression of BGP protein in periodontal tissue of periodontitis group, hypoxia group and combined group (P<0.05). The levels of MMP-2 in the combined group were significantly higher than those in the periodontitis group and hypoxia group (P<0.05), and the level of BGP protein in the periodontal tissue of the combined group was significantly lower than that of the normal group Periodontitis group and hypoxia group (P<0.05). Conclusion The expression of MMP-2 and inflammatory factor was increased in periodontal tissues of rats with periodontitis, and the expression of BGP was significantly decreased. The hypoxic condition could promote the aggravation of periodontitis.
