缺锌对NPP小鼠学习记忆功能的影响

中文摘要:目的 探究低锌处理对第5腰椎(L5)脊神经结扎横切所致神经病理性疼痛(NPP)模型小鼠的学习记忆和行为学的影响。方法 选取30只SD小鼠,采用数字表法随机分为3组(n=10):对照组、模型组(NPP)及低锌NPP组(LZn-NPP)。3组术后分别在3、7、14天进行机械痛阈检测,14天后进行Morris水迷宫实验测量小鼠空间学习记忆能力。水迷宫实验结束后,甲苯胺蓝染色法观察海马组织形态变化。利用原子吸收光谱方法测定血清锌离子,来观察低锌处理对脊神经结扎致痛模型小鼠学习记忆和行为学的影响。结果 对照组、NPP组、LZn-NPP组机械痛阈值逐渐降低(P<0.05)。Morris水迷宫实验中,NPP组21.88±11.67m/s,LZn-NPP组24.80±8.32m/s及对照组9.62±3.90m/s,隐藏平台潜伏期逐渐缩短(P<0.05)。甲苯胺蓝染色法结果显示,NPP组海马区细胞数量减少,LZn-NPP组海马区细胞数量明显减少,对照组海马区细胞形态完整。原子吸收光谱检测结果显示,对照组、NPP组及LZn-NPP组锌含量逐渐减少。结论 低锌处理可以使神经病理性疼痛小鼠的学习记忆能力下降,锌离子对疼痛模型的学习记忆功能有显著影响。

Effects of Zinc Deficiency on Learning and Memory Functions in Mice
Abstract:Objective To investigate the impact of low zinc treatment of learning,memory and ethology in mice with neuropathic pain model which caused by spinal nerve ligation. Methods Thirty mice were divided into 3 groups(n=10) in random number table way:Control group,NPP group and LZn-NPP group. On the 3rd, 7th and 14th days, the mechanical pain threshold was detected of three groups. On the 14th days days,we measured the learning and memory ability of three groups mice with Morris water maze experiment.When the water maze experiment is over,we observed the morphological varieties of hippocampal tissue by toluidine blue stains.We measured the quantity of Zn2+ in blood serum by using atomic absorption spectrometry technology.We investigated the impact of low zinc treatment of learning,memory and ethology in mice with neuropathic pain model which caused by spinal nerve ligation. Results Mechanical pain threshold decreased gradually in Control group,NPP group and LZn-NPP group(P<0.05).In morris water maze experiment,NPP group 21.88±11.67m/s,LZn-NPP group 24.80±8.32m/s and Control group 9.62±3.90m/s,Hidden platform latency decreased gradually(P<0.05).The result of toluidine blue stains shows the number of cells in the hippocampus of the NPP group decreased. The number of cells in the hippocampus of the LZn-NPP group decreased significantly, and the cell morphology of the hippocampus in the Control group was intact. Atomic Absorption Spectrometry showed that the zinc content decreased gradually in Control group, NPP group and LZn-NPP group. Conclusion Low zinc treatment can reduce the learning and memory ability of neuropathic pain mice, and zinc ion has a significant influence on the learning and memory function of pain model.
