三七皂苷Fc对高糖损伤血管内皮细胞保护作用

中文摘要:目的 探讨三七皂苷Fc对体外高糖诱导大鼠血管内皮细胞损伤的保护作用。方法 体外培养大鼠血管内皮细胞(RAOEC),加入30mmol/L D-葡萄糖造成细胞损伤。加入不同浓度三七皂苷Fc进行干预,采用CCK-8法测定细胞活性,筛选出最适用药浓度。将细胞均匀接种于6孔板内,分为4组,即正常对照组、高糖组、正常对照+Fc组、高糖+Fc组,用Hochest33342荧光染色法对各实验组进行细胞凋亡的检测;采用细胞划痕实验检测各组细胞的伤口修复情况;采用Western blot法观察各组过氧化物酶体增殖物激活受体-γ(PPAR-γ)的蛋白表达。结果 高糖+Fc组内皮细胞凋亡率明显低于高糖组,而修复率却明显高于高糖组,差异均有统计学意义(P<0.05);高糖+Fc组PPAR-γ蛋白表达量明显高于高糖组,差异有统计学意义(P<0.05)。结论 三七皂苷Fc可抑制高糖引起的血管内皮细胞凋亡,并促进高糖状态下血管内皮细胞的增殖修复。其作用机制可能与三七皂苷Fc促进高糖状态下血管内皮细胞的PPAR-γ蛋白表达有关。

Protective Effect of Notoginsenoside Fc on Vascular Endothelial Cell Against Injury Induced by High Glucose
Abstract:Objective To investigate the protective effect of Notoginsenoside Fc on injure of rat vascular endothelial cells induced by high glucose in vitro. Methods Rat vascular endothelial cells (RAOEC) were cultured in vitro, and 30mM glucose was used to induce cell injury. Notoginsenoside Fc was added to the cells for intervention. Cell viability was determined by CCK-8 assay, and the most suitable concentration was selected. The cells were cultured in 6 well plates, divided into 4 groups:normal control group, high glucose group, normal control +Fc group, and high glucose +Fc group. Hochest33342 fluorescence stain were used to detect the cell apoptosis among every group; and cell scratch test were used to observe wound repairing.The PPAR-γ expression were measured by Western blot. Results Compared with high glucose group, the apoptosis rate of endothelial cells in high glucose +Fc group was significantly lower, and the repair rate is significantly higher, the differences were statistically significant (P<0.05). The expression of PPAR-γ in high glucose +Fc group was significantly higher than that of high glucose group, the difference was statistically significant (P<0.05). Conclusion Notoginsenoside Fc can inhibit the apoptosis and promote the proliferation of vascular endothelial cells under high glucose condition. The mechanism may be related to the promotion of PPAR-γ expression due to the treat with Notoginsenoside Fc.
