丝素/明胶组织工程支架的生物相容性初探

中文摘要:目的 初步研究丝素/明胶(silk fibroin/gelatin,SFG)支架的生物相容性。方法 利用冷冻干燥法制备SF/G支架,将1×105/ml的第3代人牙周膜干细胞(human periodontal ligament stem cells,hPDLSCs)接种于SF/G支架上,设置对照组(人牙周膜干细胞悬液),通过MTT试验测试两组人牙周膜干细胞第1、3、5、7天的吸光度值,利用细胞扫描电镜观察第5天和第7天两组细胞的生长增殖状况。结果 MTT显示接种于SF/G支架上的hPDLSCs的吸光度值在第1、3、5、7天分别为0.053、0.113、0.197、0.399,对照组分别为0.069、0.105、0.176、0.283。细胞扫描电镜显示第5、7天的hPDLSCs在SF/G支架上相互融合,分泌大量细胞外基质,细胞触须几乎铺满整个支架表面,并且有朝向支架空隙内部生长的趋势。结论 冷冻干燥法制备的SF/G支架能够促使细胞在其上增殖生长,具有一定的生物相容性,在牙周组织工程领域具有一定的应用潜力。

Biocompatibility Preliminary Investigation of Silk Fibroin/Gelatin Tissue Engineering Scaffolds
Abstract:Objective To examine the biocompatibility of silk fibroin(SF)/Gelatin,(G)scaffolds. Methods SF/G scaffolds were made by freezing-dry method.Passages 3 human periodontal ligament stem cells at a density of 1×105/ml were seeded on the scaffolds to evaluate its cellular biocompatibility by MTT assay and observed by scanning electron microscope. Results The MTT results showed that the human periodontal ligament stem cells seeded on the silk fibroin/Gelatin scaffolds had higher A values compared with blank control group,and the 5th and 7th days cell scanning electron microscopy figures also showed the human periodontal ligament stem cells grew well on the scaffolds. Conclusion SF/G scaffolds has good biocompatibility and application potential in periodontal tissue engineering.
