中文摘要:目的 利用噬菌体七肽库,筛选出与小细胞肺癌细胞系H446细胞特异性结合的多肽。 方法 利用噬菌体展示技术和小细胞肺癌细胞系H446细胞,经过多次3轮筛选,每次挑取12个噬菌体单克隆进行DNA测序,分析得到多肽序列,并利用免疫组织化学技术验证多肽的特异性。 结果 多肽AF(AGALHQF)与小细胞肺癌病理组织有较高的亲和性。 结论 噬菌体展示技术筛选的多肽AF可望为小细胞肺癌靶向治疗提供新的方向。 |

Screening of Small Cell Lung Cancer Cells Specific Polypeptide from a Phage Display Peptide Library |

Abstract:Objective To obtain the polypeptides, which specifically recognize small cell lung cancer cells (SCLCs) from peptide libraries. Methods After several times of three rounds of biopanning by applying phage display technology in SCLCs H446 cells, we selected twelve clones to extract DNA, and then these DNA were sequenced and deduced to amine acid sequence. Immunohistochemical method was used to identify the specificity of these polypeptides. Results Polypeptide AF specifically recognized SCLC tissue section. Conclusion Polypeptides AF, obtained by phage displayed technology, potentially provides new strategy in the targeting treatment of small cell lung cancer. |
