| 中文摘要:目的 观察疏肝化痰祛瘀方含药血清对人乳腺癌细胞MCF-7增殖能力的调节。方法 应用疏肝化痰祛瘀方中药汤剂给予大鼠灌胃,取大鼠腹主动脉血制备含药血清。将人乳腺癌细胞MCF-7在体外进行传代培养,分3组进行干预其生长,分别为空白组、对照组(加入表阿霉素)、实验组(加入疏肝化痰祛瘀方含药血清)。3组分别培养24、48及72h,检测细胞增殖及凋亡情况,对比各组不同作用时间细胞增殖抑制率及凋亡率。结果 疏肝化痰祛瘀方对MCF-7细胞体外增殖具有一定的抑制作用,随着作用时间的延长,抑制作用逐渐增强(P<0.05),与表阿霉素组作用相似;疏肝化痰祛瘀方可诱导MCF-7细胞凋亡,与作用时间呈正相关(P<0.05)。结论 疏肝化痰祛瘀方含药血清具有抑制人乳腺癌细胞MCF-7增殖与诱导其凋亡的作用,与作用时间呈正相关,提示该方剂发挥临床治疗乳腺癌的疗效机制所在。 |

| Mechanism of Shuganhuatanquyufang on Regulating MCF-7 Cell Proliferation |

| Abstract:Objective To investigate the mechanism of Shuganhuatanquyufang on regulating human adenocarcinoma cell (MCF-7) proliferation. Methods Serum containing drug was obtained from the blood achieved in rodent aorta abdominalis after Shuganhuatanquyufang gavage. In vitro MCF-7 cell were separated into control group, positive control group (with pharmorubicin) and drug treated group (with Serum containing Shuganhuatanquyufang).After 24, 48, 72h the cell proliferation and apoptosis ratio were examined. Results The suppressive effect on MCF-7 proliferation played a time depended manner(P<0.05). The condition was similar in positive control group and drug treated group. The MCF-7 cell apoptosis ratio in drug treated group also played a time depended manner(P<0.05). Conclusion Serum containing Shuganhuatanquyufang drug could suppress MCF-7 cell proliferation and induce apoptosis in a time depended manner, which might be the possible mechanism of this prescription on treating human adenocarcinoma in clinic. |
