中文摘要:目的 研究自噬在晚期糖基化产物(AGEs)诱导的平滑肌细胞增殖和迁移中的具体作用。方法 用AGEs处理血管平滑肌细胞后,Western blot法观察自噬相关蛋白LC3-Ⅱ和SQSTM1/p62的表达变化;MTT实验检测细胞增殖水平;细胞小室实验测定细胞迁移的能力。结果 AGEs刺激血管平滑肌细胞后,自噬相关蛋白LC3-Ⅱ表达增加,SQSTM1/p62表达减少。与对照组相比,AGEs(100μg/ml)处理组显著增强血管平滑肌细胞的增殖和迁移能力。但是,使用自噬抑制剂3-MA预处理血管平滑肌细胞可减弱这种现象。结论 本研究证实AGEs诱导的自噬可增强AGEs诱导的血管平滑肌细胞的增殖和迁移能力。 |

Role of Autophagy in Advanced Glycation End Product-induced Proliferation and Migration in Rat Vascular Smooth Muscle Cells |

Abstract:Objective To identify the role of autophagy in advanced glycation end product-induced proliferation and migration in rat vascular smooth muscle cells. Methods After cultured rat vascular smooth muscle cells (VSMCs) were treated with advanced glycation end products (AGEs), autophagy-associated protein LC3-Ⅰ/Ⅱ and SQSTM1/p62 was detected by Western blot. The cell proliferation was quantified using the MTT assay. The cell migration was evaluated by Transwell migration assay. Results After cultured VSMCs were treated with AGEs. The level of LC3-Ⅱ was up-regulated whereas SQSTM1/p62 was decreased. AGEs (100μg/ml) increased the proliferation and migration potential significantly as compared to the control group in VSMCs. However, pretreating cells with 3-MA, an autophagy inhibitor, could attenuate these effects. Conclusion Our study demonstrated that AGE-induced autophagy accelerated AGE-stimulated proliferation and migration in vascular smooth muscle cells. |
