中文摘要:目的 探讨Patched1(Ptch1)、融合阻抑蛋白Su(Fu)基因在胰腺癌中的表达及其与胰腺癌发生、发展的关系.方法 采用RT-PCR、Western blot法检测56例胰腺癌组织中Ptch1、Su(Fu)基因和蛋白的表达情况,并分析其与胰腺癌临床病理参数之间的关系.结果 RT-PCR检测结果显示,Ptch1、Su(Fu) mRNA在胰腺癌中相对表达量分别为0.413±0.045、0.376±0.053,在癌旁组织中分别为0.203±0.013、0.214±0.062(P<0.05).Western blot法检测结果显示,Ptch1、Su(Fu) 蛋白在胰腺癌中的相对表达量分别为0.724±0.021、0.625±0.075,在胰腺癌旁组织中分别为0.516±0.098、0.413±0.051.胰腺癌组织中Ptch1、Su(Fu) mRNA与蛋白表达均高于癌旁组织.胰腺癌组织中Ptch1、Su(Fu) mRNA与蛋白表达与胰腺癌的分化程度和TNM分期明显相关(P<0.05),与患者年龄、性别、肿瘤直径、侵犯血管和神经无明显相关(P>0.05).结论 胰腺癌组织中Ptch1、Su(Fu)蛋白呈高表达,Ptch1、Su(Fu)蛋白的高表达可能与胰腺癌的发生及发展有关. |

Abstract:Objective To investigate the relationship between Patched1(Ptch1),Suppressor of fused expression and the development of human pancreatic cancer. Methods RT-PCR and Western blot were used to determine the expression of Ptch1,Su(Fu) in 56 cases of pancreatic cancer quantitatively.The relationship between the expression of Ptch1,Su(Fu) and the clinicopathological features of pancreatic cancer was analyzed. Results The expression of Ptch1,Su(Fu) mRNA in pancreatic cancer was 0.413±0.045, 0.376±0.053,and in pancreatic cancer adjacent tissue was 0.203±0.013,0.214±0.062 by RT-PCR detected(P<0.05).The expression of Ptch1,Su(Fu) protein in pancreatic cancer was 0.724±0.021,0.625±0.075,and in pancreatic cancer adjacent tissue was 0.516±0.098,0.413±0.051 by Western blot detected(P<0.05).The expression of Ptch1,Su(Fu) mRNA and protein in pancreatic cancer tissue were both higher than pancreatic cancer adjacent tissue.The expression of Ptch1,Su(Fu) was significant associated with differentitation and TNM grade(P<0.05),not age, gender,tumorous size and invasion to nerves(P>0.05). Conclusion The Ptch1,Su(Fu) overexpressed in pancreatic cancer tissues and its abnormal expression may be involved in the oncogenesis and development of pancreatic cancer. |
